miR99-a在早期自然流产患者中表达情况及其与孕期激素水平的相关性

         

摘要

目的:探讨miR-99a在早期自然流产中表达情况及其与孕期激素水平的相关性。方法通过病例-对照研究,选取2014年1月1日至2015年12月31日就诊于南京医科大学附属淮安第一医院妇产科,经确诊为早期自然流产的孕妇62例为早期自然流产组,同期非医学原因选择流产的正常妊娠孕妇83例为对照组。采用酶联免疫吸附测定法对入选样本的激素水平进行检测;利用实时定量聚合酶链反应(qRT-PCR)检测miR-99a在病例与对照组中的表达情况;利用偏相关分析方法,对血清激素水平及miR-99a的表达水平进行相关性分析。结果激素水平分析发现,早期自然流产组血清激素β-人绒毛膜促性腺激素(β-HCG)、孕酮、雌激素水平均低于对照组[(4774±1704) mU/L比(10326±4032) mU/L、9.7(6.8,15.2)μg/L比19.7(16.6,22.9)μg/L、334.2(213.3,811.7) ng/L比909.2(720.8,1135.1) ng/L],差异有统计学意义(P<0.01);早期自然流产组miR-99a表达水平高于对照组(1.60±0.25比0.60±0.11),差异有统计学意义(P<0.01);偏相关分析提示,血清激素β-HCG、孕酮、雌激素水平均与miR-99a的表达水平呈负相关(r=-0.59,-0.43,-0.55,P<0.01)。结论 miR-99a在早期自然流产患者血清中高表达,其表达水平与孕期激素水平存在一定的关联,可望作为新的候选分子诊断标志物应用于自然流产诊断。%Objective To study the expression of miRNA99-a in early spontaneous abortion and its rela-tionship with hormone levels during pregnancy.Methods By a case-control study,62 cases of early sponta-neous abortion in Department of Gynecology and Obstetrics of the First Affiliated Hospital of Nanjing Medical University from Jan.2014 to Dec.31st 2015 were included as an early spontaneous abortion group,and 83 cases of normal pregnancy women for abortion due to non-medical reasons were included as a normal control group.Enzyme-linked immunosorbent assay was used to test the hormone levels;quantitative real-time poly-merase chain reaction(qRT-PCR) was used to detect miR-99a expression;partial correlation analysis method was used to analyzed the correlation between serum hormone levels and miR-99a expression level.Results The average levels of serum hormone β-human chorionic gonadotropin (β-HCG ) , progesterone ( PROG ) , estradiol (E2) of the early spontaneous abortion group were lower than the normal control group[(4774 ± 1704) mU/L vs (10 326 ±4032) mU/L,9.7(6.8,15.2) μg/L vs 19.7(16.6,22.9) μg/L,334.2 (213.3,811.7) ng/L vs 909.2(720.8,1135.1) ng/L],the difference were statistically significant (P<0.01);the expression level of miRNA-99a of the early spontaneous abortion group was higher than the nor-mal control group(1.60 ±0.25 vs 0.60 ±0.11),the difference was statistically significant(P<0.01);par-tial correlation analysis indicated that the level of serum hormone β-HCG,PROG,E2 were negatively correla-ted with the expression of miRNA99-a (r=-0.59,-0.43,-0.55,P<0.01).Conclusion miRNA-99a is highly expressed in patients of early spontaneous abortion , and is correlated with the gestational hormone levels,which is expected to be used as a new clinical marker in spontaneous abortion diagnosis.
