他汀类药物临床多效性研究进展

摘要

他汀类药物被认为是降低血清胆固醇水平最有效的药物.他汀类药物除脂质调节作用外的其他非脂质调节功能的多效性可使不同疾病的患者受益.现发现他汀类药物最重要的非脂质调节作用包括抗炎、抗氧化、抗栓、改善内皮功能、减轻血管重构、免疫调节及神经保护等.在上述生物学进程中,R孙o/Rho相关卷曲蛋白激酶、Ras相关C3肉毒毒素底物、过氧化物酶体增殖物激活受体等信号通路可视为他汀类药物的多效性的主要分子生物学机制.%Statins have been viewed as the most effective drugs to reduce cholesterol serum level.Recent studies have demonstrated non-lipid modifiable pleiotropic effects of statins,which could benefit patients with different diseases.The most important non-lipid modifiable effects of statins include anti-inflammation,antioxidant,antithrombosis,improving endothelial dysfunction,attenuating vascular remodeling,immunomedulation,neuroprotection,etc.In particular,Ras homolog family members(Rho)/Rho associated coiled coil-containing protein kinases,Ras-related C3 botulinum toxin substrate and peroxisome proliferator activated receptors signaling pathway can be regarded as the principle molecular mechanisms of the pleiotropic effects of statins.
