构建应用于生物学研究的组织工程肌腱

         

摘要

目的:构建应用于生物学研究的组织工程肌腱,为体外研究中检测胶原纤维的形态结构提供可行的方法手段。方法:培养大鼠肌腱细胞,构建无支架的组织工程肌腱,并检测其光镜结构和电镜结构。结果:构建的组织工程肌腱表面光滑,结构紧密,细胞生存良好,细胞外基质分泌丰富。具有波浪形的胶原纤维,胶原纤维排列规则,直径大小相对均一,但小于正常肌腱纤维直径。结论:本研究构建的组织工程肌腱不含支架材料,结构与损伤肌腱类似,可以检测多种生物学指标,适于肌腱损伤修复的体外研究。%Objective:To Construct tissue-engineered tendon for biological assay to provide practical methods and means for detecting the morphology of collagen fiber in in-vitro study.Method:Rat tendon cells were cultured and were used to construct scaffold-free tissue-engineered tendon. The structure was detected by light microscope and electron microscope.Result:The tissue engineered-tendon with living cells and collagen matrix has a smooth surface and compact structure. It has bundles of highly crimped fibers and a more uniform distribution of smaller diameter collagen fibrils than native tissue.Conclusion:This tissue-engineered tendon is scaffold-free and has a similar structure with injured tendon. It can be used for many biological assays and is applicable for studying tendon injury in vitro.
