氟化物联合五倍子水提取物对变形链球菌影响的体外研究

     

摘要

Objective:To study the effects of combined application of fluoride and the gallnut water extract on the growth and acidogenic metabolism of Streptococcus mutans(S.mutans).Method:10 mL of sterile culture tube 120 were selected and divided into 12 groups,1 group was control group,and 11 groups were experimental group,each group of 10. The experimental group standard S. mutans were cultured in vitro in the TS medium which contained 5%sucrose and different concentration of NaF(0.01%,0.02%)and/or gallnut water extract(0.25,0.5,1 mg/mL),and the medium without neither NaF nor gallnut water extract act as control group. The optical density(OD)value of the medium and pH change before and after the culture was determined. Result:Compared with the control group,the experimental group S.mutans growth was obviously inhibited,and with the increase of amount of sodium fluoride and gallnut water extract concentration of culture,the number of S.mutans showed a concentration dependent decrease;with the increase of sodium fluoride and the culture medium of gallnut water extract concentration,pH changes of medium of gradually reduced,NaF combined with gallnut water extract showed no obvious synergistic effect. Conclusion:NaF and gallnut water extract could significantly inhibit the growth and acidogenic metabolism of S. mutans. No obvious synergistic effect was observed while the NaF combined with gallnut water extract.%目的:研究氟化物与五倍子水提取物联合应用对变形链球菌生长和代谢的影响。方法:取10 mL的无菌培养管120支,分为12组,对照组1组,实验组11组,每组各10支。对照组中含无菌去离子水,实验组中含有不同浓度氟化钠(0.01%、0.02%)、五倍子水提取物(0.25、0.5、1 mg/mL)及氟化钠与五倍子联合应用的培养基内培养变形链球菌标准菌株,检测培养前后两组培养基光密度值及pH的变化。结果:与对照组相比,各实验组变链菌的生长受到明显抑制,同时各组培养基的pH变化也逐渐减小,氟化钠与五倍子联合应用时未发现明显的协同效应。结论:氟化钠及五倍子水提取物均可显著抑制变形链球菌的生长及产酸代谢活动,两者联合应用时未见明显的协同效应。
