MMP-2、MMP-9在口腔黏膜白斑中表达的研究

Objective:To investigate the expression of matrix metalloproteinase 2,9 in oral mucosa, hyperplasia and dysplasia and to discuss the correlation of MMP-2,9 with oral leukoplakia.Method:The statistical evaluation was used to analyze immunohistochemical results.The study was carried out on clinically and histologically proven 15 cases of normal mucosa and 31 cases of hyperplasia and 35 cases of dysplasia using anti-MMP-2 and anti-MMP-9 antibodies. Result:MMP-2,9 are both present in cytoplasm. In epithelium, the positive rates of MMP-2 and MMP-9 in normal oral mucosa,hyperplasia and dysplasia was 20.0%,22.6%,34.3% and 6.7%,9.7%,34.3%,respectively.Under epithelium,in fibroblasts,the positive rates of MMP-2 and MMP-9 in above three groups was 0,12.9%,14.3% and 6.7%,25.8%,80.0%,respectively.There was no significant difference in the expression of MMP-2 between three groups(P>0.05),but the expression of MMP-9 was higher in dysplasia than in normal oral mucosa and hyperplasia (P<0.05).Conclusion:MMP-9 maybe plays a role in the occurrence and development of leukoplakia.%目的：检测基质金属蛋白酶2、9（matrix metalloproteinase 2、9，MMP-2、9）在口腔正常黏膜、白斑上皮单纯性增生及异常增生中的表达，探讨其与白斑不同增生程度间发生发展的关系。方法：选取正常口腔黏膜15例，白斑单纯性增生31例，白斑异常增生35例，应用MMP-2、MMP-9抗体行免疫组化染色并进行统计学分析。结果：MMP-2、MMP-9均表达在细胞浆，在上皮内两者的阳性率在上述三组中分别为20.0%、22.6%、34.3%和6.7%、9.7%、34.3%，在上皮下成纤维细胞中两者的阳性率在上述三组中分别为0、12.9%、14.3%和6.7%、25.8%、80.0%。MMP-2在上皮内、成纤维细胞的阳性表达在三组间差异无统计学意义（P>0.05）。MMP-9在上皮内及上皮下成纤维细胞的阳性表达在白斑异常增生组中明显高于正常组织和白斑单纯增生组（P<0.05）。结论：MMP-9可能参与了白斑发生发展的过程。
