免疫磁微球在肿瘤细胞中的提取应用

             

摘要

Objective:IMMS is a new method of collecting body fluid tumor cells,and it is used to find out whether there is no shedding of tumor cells in the body fluid of the patients.Method:The tumor cells were collected and compared with the conventional sedimentation centrifugation method.Body fluids were collected from 85 patients with tumor and 53 patients with non tumor,in the same volume of body fluids,immune magnetic microsphere suspension and 138 patients with body fluids(urine sediment,ascites,peritoneal effusion,pleural effusion,nipple discharge)mixture,the collected suspicious cells in conventional microscopy,and separated the microscopic examination of the 138 patients with body fluids sediment precipitation centrifugal,compared the two kinds of mirror inspection results.Result:The positive rate of immune magnetic microsphere detection method in non tumor patients was 1.9%(1/53),the positive rate of precipitation centrifugation test method was 0(0/53).The positive rate of immune magnetic microsphere detection in patients with hepatocellular carcinoma was 91.6%(11/12),the positive rate of precipitation centrifugation test method was 33.3%(4/12).The positive rate of immune magnetic microsphere detection in patients with colon cancer was 88.9%(24/27),the positive rate of precipitation centrifugation test method was 37.0%(10/27).The positive rate of the detection method of the immune magnetic microsphere in patients with lung cancer was 80.0%(12/15),the positive rate of precipitation centrifugation test method was 40.0%(6/15).The positive rate of immune magnetic microsphere detection in patients with breast cancer was 90.3%(28/31),the positive rate of precipitation centrifugation test method was 45.2%(14/31).In patients with non tumor and tumor patients,the positive detection rate of the immune magnetic microsphere detection method were higher than those in precipitation centrifugation test method(P<0.05). Conclusion:Immune magnetic microsphere detection method easily ruled out normal cells mixed in body fluids in the sediment,broken cells and non cellular components,excluding factors interference microscopy,increase the screening concentration of positive cells in efforts to improve the positive detection rate,has a great effect on tumor diagnosis,pathological diagnosis,clinical tumor cytology more effective and convenient method.%目的:免疫磁微球是现代新颖的收集体液瘤细胞方法,用以发现受检患者体液中有无脱落肿瘤细胞。方法:分析比较免疫磁微球法与常规沉淀离心法收集肿瘤细胞。收集85例肿瘤患者和53例非肿瘤患者的体液,在同等体积的体液中,用免疫磁微球悬液与138例患者体液沉渣(尿液、腹水、腹腔积液、胸腔积液、乳头溢液)相混合,将收集到的可疑细胞进行常规镜检;并将138例患者体液沉渣进行沉淀离心镜检,比较两种镜检结果。结果:非肿瘤患者免疫磁微球检测方法阳性率为1.9%(1/53),沉淀离心检测方法阳性率为0(0/53)。肝癌患者免疫磁微球检测方法阳性率为91.6%(11/12),沉淀离心检测方法阳性率为33.3%(4/12);结肠癌患者免疫磁微球检测方法阳性率为88.9%(24/27),沉淀离心检测方法阳性率为37.0%(10/27);肺癌患者免疫磁微球检测方法阳性率为80.0%(12/15),沉淀离心检测方法阳性率为40.0%(6/15);乳腺癌患者免疫磁微球检测方法阳性率为90.3%(28/31),沉淀离心检测方法阳性率为45.2%(14/31)。在非肿瘤患者和肿瘤患者中,免疫磁微球检测方法的肿瘤阳性检出率均高于常规沉淀离心检测方法(P<0.05)。结论:免疫磁微球检测方法轻易排除了混杂在体液沉渣中的正常细胞、破碎细胞和非细胞成分,排除干扰镜检的诸多因素,加大了筛选浓聚阳性细胞的力度,提高检测的阳性率,对肿瘤诊断有很大的效用,为临床肿瘤细胞学病理确诊提供更有效、便捷的方法。
