布渣叶总生物碱在高脂血症大鼠中的作用机制分析

             

摘要

目的:探讨布渣叶总生物碱在高脂血症大鼠中的作用机制。方法:提取、分离纯化布渣叶总生物碱,高脂饲料造模,根据不同灌胃方式将Wistar大鼠随机分为对照组、阳性对照组、模型对照组和布渣叶总生物碱的低、中、高剂量组,每组各10只,测定总胆固醇(TC)、甘油三脂(TG)、脂肪酸合成酶(FAS)与肉毒碱棕榈酰转移酶-1(CPT-1)水平。结果:阳性对照组、模型对照组、布渣叶总生物碱组血清与肝脏匀浆液TC、TG水平均明显低于对照组(P<0.05),布渣叶总生物碱组上述指标均明显低于阳性对照组(P<0.05),随着布渣叶总生物碱的剂量的不断增加,血清与肝脏匀浆液TC、TG水平呈下降趋势(P<0.05)。阳性对照组、模型对照组、布渣叶总生物碱组FAS水平均明显低于对照组, CPT-1水平均明显高于对照组(P<0.05),布渣叶总生物碱组FAS水平明显低于阳性对照组,CPT-1水平明显高于阳性对照组(P<0.05),随着布渣叶总生物碱的剂量的不断增加,FAS水平呈下降趋势,CPT-1水平呈增高趋势(P<0.05)。结论:布渣叶总生物碱通过降低FAS与增高CPT-1含量从而发挥对高脂血症的治疗作用。%Objective:To study mechanism analysis of Paniculata Total Alkaloids in hyperlipidemia rats. Method:Paniculata Total Alkaloids were extracted,separated and purified,Wistar rats model were made by high fat feed,all were randomly divided into control group,positive control group,model control group and Paniculata Total Alkaloids low,medium and high dose group according to different lavage way,each for 10 cases,total cholesterol(TC),triglycerides(TG),fatty acid synthase (FAS) and carnitine palmitoyl transferase 1 (CPT) levels were determined.Result:Serum and liver lipids TC,TG levels of positive control group,model control group,Paniculata Total Alkaloids group were significantly lower than control group(P<0.05),the above index of Paniculata Total Alkaloids group were significantly lower than positive control group(P<0.05),with the dose of Paniculata Total Alkaloids increasing,serum and liver lipids TC,TG levels were declined(P<0.05).FAS level of positive control group, model control group,Paniculata Total Alkaloids group were significantly lower than control group,CPT-1 level were obviously higher than control group,FAS levels of Paniculata Total Alkaloids group significantly lower than positive control group,CPT-1 level were obviously higher than positive control group (P<0.05),with the doses of Paniculata Total Alkaloids increasing,FAS level were declined,CPT-1 level were increased(P<0.05).Conclusion:Paniculata Total Alkaloids plays a therapic role for hyperlipemia by lowering FAS and increasing CPT-1 content.
