瓯柑果皮挥发油成分的提取及GC/MS分析

         

摘要

Essential oil in skin of Citrus suavissima was extracted by supercritical CO_2 extraction and steam distillation, and isolated and identified by GC/MS analysis. Their chemical compositions and relative contents were determined by area normalization. Chromatographic conditions were: capillary column HP-5 (30 m×0.25 mm, 0.25 μm). The carrying gas was helium, the flow rate was tmL/minute and the temperature was in rate of 5 ℃/minute from 60℃(hold 2 minutes) to 240℃(kept 30 minutes) Split injection with ratio of 50:1. Injection temperature of 280℃. 32 and 31 volatile compounds were identified from chemical materials by supercritical CO_2 extraction and by steam distillation, accounting for over 93.42% and 90.86% of total essential oil fraction. The results showed that supercritical CO_2 extraction method had the advantages of faster extraction without heating, lower energy consumption, higher extraction efficiency than steam distillation.%采用超临界CO_2萃取法及常规水蒸汽蒸馏法从瓯柑果皮中提取挥发油,利用气相色谱-质谱联用技术对挥发油化学成分进行分离鉴定,采用峰面积归一化法测定其相对含量.色谱条件:HP-5弹性石英毛细管色谱柱(30m×0.25 mm,0.25 μm);升温程序:初始温度60℃,保持2 min,然后以5℃/min的速度升至240℃并保持30 min;分流进样,分流比50:1;进样口温度280℃.结果采用超临界CO_2萃取共鉴定出32种成分,所鉴定的组分占总峰面积的93.42%;采用水蒸汽蒸馏法提取共鉴定出31种成分,所鉴定的组分占总峰面积的90.86%.超临界CO_2萃取法和水蒸汽蒸馏法所提得的挥发油化学成分基本一致.超临界CO_2萃取法比传统的水蒸汽蒸馏法具有提取快速、无需加热、能耗低、有效成分的萃取率较高等优点.
