白藜三醇通过HIF途径对快速心房起搏猪心房结构重构的抑制作用

     

摘要

Objective To explore the effect and mechanism of resveratral ( Res ) on structural remodeling in right a-trium of pigs induced by right atrial tachypacing. Methods A porcine model of chronic atrial fibrillation was established by rapid right atrial tachypacing. The pigs were divided into three groups, sham operation ( sham ) group, rapid atrial pacing ( RAP ) group and RAP + Res treated group ( Res group ). Two weeks later we harvested myocardial tissues of right atrium. The deposition of collagen in cardiac tissue was tested by Masson trichrome staining. The expression of HIF - la,VEGF and MMP - 9 and the corresponding mRNAs in the right atrium were tested by Western blot and RT - PCR. Results HIF - lα, VEGF and MMP - 9 expression were significantly higher in RAP group than in sham group ( P < 0. 05 ) and Res group ( P <0. 05 ). Conclusion Res may suppress myocardial fibrosis by down - regulating HIF - lα to inhibit structural remodeling of atrium.%目的 探讨白藜三醇(resveratrol,Res)对快速心房起搏引起的猪右心房结构重构的影响以及作用机制.方法 快速右心房起搏建立猪慢性心房纤颤模型.实验分为假手术(sham)组、快速心房起搏(RAP)组和RAP+白藜三醇干预组(Res组).2周后取右心房心肌组织,Masson三色染色法比较各组心肌间质胶原沉积,Western blot和RT-PCR法检测各组心肌组织中低氧诱导因子-1α(HIF-1α)、血管内皮细胞生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)蛋白和mRNA表达水平.结果 RAP组HIF-1α、VEGF、MMP-9蛋白及mRNA含量明显高于sham组(P<0.05)和Res组(P<0.05).结论 白藜三醇可能通过降低HIF-1α的表达这一途径来改善猪心房纤颤心肌纤维化,减轻心房结构重构.
