胰岛素样生长因子-1诱导新生小鼠海马源性神经干细胞增殖分化的研究

         

摘要

目的 探讨胰岛素样生长因子-1(IGF-1)对薪生小鼠海马源性神经干细胞(NSCs)增殖和分化的影响.方法 通过取新生C57BL/6J小鼠海马组织,机械分离,体外培养NSCs,特异性标志蛋白Nestin免疫荧光鉴定.在细胞培养液中加入100 μg/L的IGF-1记为IGF-1组,同时设立正常细胞对照组.CCK-8试剂盒检测细胞增殖情况,GFAP、β3 Tubulin细胞免疫化学检测细胞分化情况.结果 体外成功培养NSCs,特异性蛋白Nestin具有表达.IGF-1组NSCs较对照组增殖显著,且IGF-1组细胞具有显著的迁移和分化能力.细胞免疫荧光鉴定,IGF-1可促进细胞分化为星形胶质细胞和神经元(P<0.05).结论 IGF-1能够促进体外培养的新生小鼠海马源性NSCs增殖,并诱导NSCs向星形胶质细胞和神经元分化.%Objective To detect the effects of insulin - like growth factor - 1 (IGF - 1) on the proliferation and dif ferentiation of cultured hippocampal neural stem cells (NSCs) of newborn mouse. Methods NSCs were cultured in vitro through collecting the hippocampus of newborn C57BL/6J mouse and medical separation and then which were detected with specific protein Nestin immunofluorescent staining. The cell culture medium was added 100 μg/L of IGF - 1 as IGF -1 group and the culture medium with no IGF -1 as control group. Cells proliferation was detected by CCK - 8 kit and cells differentiation was detected by GFAP and (33 tubulin immunofluorescent staining. Results Successfully cultured NSCs in vitro, and specific protein Nestin had expression. IGF - 1 group cells had more obvious proliferation, migration and differentiation compared with the control group. Immunofluorescent identification showed that IGF - 1 could promote cells to differentiate into astrocytes and neuron (P < 0. 05). Conclusion IGF - 1 can promote the proliferation of hipp ocampal NSCs of newborn mouse in vitro, and induce the differentiation of NSCs to astrocytes and neuron.
