CXCL8在结直肠癌组织中的表达及其临床意义

摘要

目的 探讨CXCL8在结直肠癌组织及癌旁组织中的表达,分析其与结直肠癌患者临床病理参数的相关性.方法 选择2014年11月至2016年11月于郑州市中心医院胃肠外科行手术切除的60例结直肠癌患者的癌组织及癌旁组织为研究对象,所有标本经病理学证实.免疫组织化学法和实时荧光定量聚合酶链反应检测CXCL8蛋白和mRNA在结直肠癌组织及癌旁组织中的表达.结果 CXCL8 mRNA在结直肠癌组织和癌旁组织中的表达分别为157.6±26.2和42.7±9.6,CXCL8 mRNA在结直肠癌组织中的表达显著高于癌旁组织(P<0.05).CXCL8蛋白在结直肠癌组织和癌旁组织中的阳性表达率分别为83.3％(50/60)和25.0％(15/60),CXCL8蛋白在结直肠癌组织中的阳性表达率高于癌旁组织(χ2=41.10,P<0.01);CXCL8在结直肠癌组织中的表达与淋巴结转移、肿瘤分期有关(P<0.05),与结直肠癌患者的年龄、性别及结直肠癌分化程度无相关性(P>0.05).结论 CXCL8在结直肠癌组织中呈高表达,且与结直肠癌患者的淋巴结转移情况、肿瘤分期有相关性.%Objective To detect the expression of CXCL8 in colorectal tumor tissues and marginal tissues,and analyze the correlation between CXCL8 and clinicopathological parameters. Methods Sixty colorectal cancer specimens and the mar-ginal tissues were collected from the colorectal cancer patients who were surgically removed in gastrointestinal surgery in Zhengzhou Central Hospital Affiliated to Zhengzhou University from November 2014 to November 2016 and were all confirmed by pathology. The expression of CXCL8 protein and mRNA in colorectal tumor tissues and marginal tissues was detected by im-munohistochemistry and real time fluorescent quantitation polymerase chain reaction respectively. Results The expression of CXCL8 mRNA in colorectal tumor tissues and marginal tissues was 157. 6 ± 26. 2 and 42. 7 ± 9. 6. The expression of CXCL8 mRNA in colorectal tumor tissues was higher than that in the marginal tissues(P < 0. 05). The positive expression rate of CXCL8 protein in the colorectal tumor tissue and marginal tissue was 83. 3％(50 / 60)and 25. 0％(15 / 60),respectively. The positive expression rate of CXCL8 in the colorectal tumor tissues was higher than that in the marginal tissues(χ2 = 41. 10,P <0. 01). The expression of CXCL8 in colorectal tumor tissues was correlated with lymph node metastasis and tumor staging(P <0. 01). The expression of CXCL8 in colorectal tumor tissues had not correlation with age,gender and differentiated degree colo-rectal tumor(P > 0. 05). Conclusion CXCL8 has high expression in colorectal cancer tissues,and it is closely related to the condition of lymph node metastasis and tumor staging in patients with colorectal cancer.
