薄层扫描法测定毛头牛蒡子中牛蒡苷和苷元的含量

         

摘要

Objective To set up the contents determination method of arctiin and arctigenin in Arctium tomentosum Mill seeds.Methods A TLCS method was used.The developer was consisted of chloroform and n-butyl alcohol of 10∶1,arctiin and arctigenin were isolated by high performance thin layer plate and scanned at 280 nm.Results The liner ranges of calibration curves were 1.020-6.120 mg/mL and 0.101-0.606 mg/mL for arctiin and arctigenin respectively.The sample recovery rate of arctiin and arctigenin were 99.71% and 98.11% respectively.Contents of arctiin and arctigenin inArctiumtomentosum Mill seeds were 93.75 mg/g and 2.05 mg/g respectively.Conclusion The method is simple,accurate,reproducible and can be used to determine the contents of arctiin and arctigenin of Arctium tomentosum mill.%目的:测定毛头牛蒡子中牛蒡苷和苷元的含量。方法以氯仿-甲醇(10∶1)为展开剂,在高效薄层板上对毛头牛蒡子中的牛蒡苷及苷元进行分离,并以280 nm为检测波长对毛头牛蒡子进行扫描测定。结果牛蒡苷和牛蒡苷元的线性范围分别为1.020~6.120、0.101~0.606μg;加样回收率分别为99.71%和98.11%,毛头牛蒡子中牛蒡苷和苷元的含量分别为93.75 mg/g 和2.05 mg/g。结论薄层扫描法简便、准确、重复性好,可用于测定毛头牛蒡子中牛蒡苷和苷元的含量。
