库页岛悬钩子化学成分预实验及急性毒性、抗疲劳实验研究

     

摘要

Objective To study the chemical components,acute toxicity & antifatigue of different extractsfromRubus sachalinensis Levl (RSL).Methods Physical and chemical identification of the water and ethnolextracts from RSL were studied by tube method.In acute toxicity experiment,60 ICR mice were randomlydivided into 6 groups,except control group mice were given equal volume of 0.5% CMC-Na,theother groups were intragastrically administrated with 0.49 g/mL crude extract,0.13 g/mL ethyl acetate extract,1.00 g/mL n-butyl alcohol extract,0.50 g/mL n-butyl alcohol extract and 1.01 g/mL water extractsof RSL respectively for two times a day lasting 14 days,and acute toxicity reaction and death situationsof mice were recorded.For studying of the RSL on anti-fatigue effect,70 ICR mice were randomly dividedinto 7 groups with equal volume of 0.5% CMC-Na,0.49 g/mL,0.25 g/mL,0.13 g/mL of crude extracts,0.13 g/mL ethyl acetate extract,n-butanol extract and water extract by intragastric administrationonce a day lasting 30 days,and compelled weight loading swimming time was determined.Results RSLextracts includes polysaccharide,glycosides,phenols,tannins,flavones,flavonol glycosides,alkaloids,organic acids,cardiac glycosides compounds.All the mice in every group survived without obvious toxicityexcept the n-butanol extract group.The experiment of compelled weight loading swimming time of themice in every group was clearly longer than that of the control group,in which the mid-dose group of crudeextract had significant difference (P <0.01).Conclusion RSL might contain polysaccharide,glycosides,phenols,tannins,flavones,flavonol glycosides,alkaloids,organic acids,cardiac glycosides componends.RSL extracts can be eaten safety with low toxicity and significantly prolong compelled weight loadingswimming time of the mice.%目的 研究库页岛悬钩子不同溶剂提取物的化学成分及其急性毒性、抗疲劳作用.方法 采用试管法对库页岛悬钩子水提取物和乙醇提取物进行化学成分预实验.急性毒性实验将60只 ICR小鼠随机分为6组,分别是空白组(等容积0.5%CMC-Na溶液)、库页岛悬钩子粗提物组(0.49 g/mL)、乙酸乙酯提取物组(0.13 g/mL)、正丁醇提取物高剂量组(1.00 g/mL)、正丁醇提取物低剂量组(0.50 g/mL)、水提取物组(1.01 g/mL),灌胃体积40 mL/kg,每天2次,每次间隔5 h,连续观察14 d,记录小鼠的急性毒性反应及死亡情况.抗疲劳实验采用70只ICR小鼠,随机分为7组,分别是空白组(等容积0.5%CMC-Na溶液)、库页岛悬钩子粗提物高剂量组(0.49 g/mL)、中剂量组(0.25 g/mL)、低剂量组(0.13 g/mL)、乙酸乙酯提取物组(0.13 g/mL)、正丁醇提取物组(0.13 g/mL)和水提取物组(0.13 g/mL),灌胃最大体积20 mL/kg,连续给药30 d,测定小鼠负重游泳时间.结果 库页岛悬钩子中含有多糖、苷类、酚类、鞣质、黄酮类、黄酮醇苷类、生物碱类、有机酸类和强心苷类化学成分.急性毒性实验表明除库页岛悬钩子正丁醇提取物组外,其余各组小鼠全部存活,毒性反应差异无统计学意义.抗疲劳实验显示,与空白组相比,各实验组小鼠负重游泳时间均有延长,其中库页岛悬钩子粗提取物中剂量组差异有统计学意义(P <0.01).结论 库页岛悬钩子中含有多糖、苷类、酚类、鞣质、黄酮类、黄酮醇苷类、生物碱类、有机酸类和强心苷类等成分,库页岛悬钩子提取物毒性较低,具有抗疲劳的作用.
