石榴皮安石榴苷提取物对人前列腺癌PC-3细胞的增殖及凋亡的影响

摘要

目的 探讨石榴皮安石榴苷提取物(PUN)对人前列腺癌PC-3细胞的增殖及细胞凋亡的影响.方法 采用细胞形态学观察与四甲基偶氮唑盐(MTT)比色法测定PUN、石榴皮多酚提取物(PPP)与氟他胺(FLU)对PC-3细胞增殖的影响.应用流式细胞仪检测石榴皮中的PUN作用48 h后诱导PC-3细胞的凋亡情况.结果 PUN与PPP对人前列腺癌PC-3细胞增殖均有明显的抑制作用,且均呈剂量与时间依赖性;PUN对PC-3细胞的抑制作用优于PPP(P<0.05),而与FLU的抑制作用比较差异无统计学意义(P>0.05);PUN与PPP可明显诱导PC-3细胞凋亡,且均呈剂量依赖性,PUN与PPP在质量浓度为400、800μg/mL时,PUN组细胞的早期凋亡率要优于PPP组,差异有统计学意义(P<0.05);PUN与FLU在质量浓度为200、800μg/mL时细胞的早期凋亡率差异无统计学意义(P>0.05).结论 PUN在体外可抑制人前列腺癌PC-3细胞的增殖,诱导细胞凋亡.随着安石榴苷含量的提高,石榴皮多酚提取物对人前列腺癌PC-3细胞的体外抑制作用也会增强.%Objective To study the anticancer activity and apoptosis effects of punicalagin of pomegranate peel (PUN)on human prostate cancer PC-3 cells.Methods Cell morphological were observed and methyl thiazolyl tetrazolium (MTT)assay were used to detect the impact on PC-3 proliferation causing by PUN, polyphenols of pomegranate peel (PPP)and flutamide (FLU).The cell apoptosis of PC-3 cells were analyzed by using flow cytometry after treated with 48 h.Results PUN and PPP significantly inhibited the cell proliferation of PC-3 cells in dose and time dependent manners.The effect of PUN was better than the effect of PPP (P <0.05).There was no significant difference between PUN and PPP (P >0.05).PUN and PPP obviously induce the apoptosis of PC-3 cells in dose dependent manners.When the mass concentration were 400 and 800 μg/mL,early apoptosis rate of PUN group is superior to that of the PPP group,and there was significant difference between the effect of inducing apoptosis of PUN and PPP (P <0.05).When the mass concentration were 200 and 800μg/mL,there was no significant difference between the effect of inducing apoptosis of PUN and FLU (P >0.05).Conclusion PUN could inhibit the proliferation and induce apoptosis of PC-3cells.When the content of punicalagin in the polyphenols extracts of pomegranate peel rise,the effect of inhibiting the proliferation and inducing apoptosis to PC-3 cells was stronger.
