氧化亚铁硫杆菌的生长特性及驯化

         

摘要

To understand the growth characteristics of Thiobacillus ferrooxidans,which is cultured in the low pH value and high temperature environment,effect of initial pH,temperature,inoculum size on the growth of Thiobacillus ferrooxidans are studied by the determination of the concentration of bacteria and Fe2+.The results show that pH 2.0,temperature 30 ℃,and 10% inoculum size are the best culture conditions of Thiobacillus ferrooxidans.The concentration of Fe2+ decreased by 97% in low pH value acclimation after 42 h culture,shortened by 12 h compared with the time before acclimation.The concentration of Fe2+ decreased by 98.4% in low pH and high temperature acclimation culture.The growth activity of Thiobacillus ferrooxidans that strains obtained after acclimation are close to which under the optimal conditions,and the purpose of the domestication is achieved.%为了解低pH值高温环境中正常生长的氧化亚铁硫杆菌的生长特性,测定其生长过程中菌液浓度和Fe2+浓度,分析初始pH、培养温度、菌液接种量对氧化亚铁硫杆菌生长的影响.以此为基础改变培养条件,驯化获取目标菌株.结果表明,氧化亚铁硫杆菌的培养条件在pH 2.0,温度30℃,菌液接种量为10%时,生长活性最佳.低pH值驯化培养,42 h后Fe2+浓度降低了97%,相比驯化前的培养时间缩短12 h;低pH值高温驯化培养,54 h后Fe2+浓度降低了98.4%.驯化培养后获得菌株的生长活性接近氧化亚铁硫杆菌在最佳条件下的活性,达到驯化目的.
