植物生长调节剂对红心杉组培苗生长及内源激素的影响

         

摘要

以红心杉优良单株组培苗为材料,采用L4(23)正交试验设计探讨不同植物生长调节剂浓度及其配比对红心杉组培苗生长及内源激素的影响。结果表明:不同植物生长调节剂对红心杉组培苗的高生长及茎段的增殖生长均有促进作用, IBA 0.3mg/L、6-BA 0.8mg/L是有利于红心杉增殖培养的植物生长调节剂浓度,当KT 1mg/L时有利于其苗高生长, KT 2mg/L时有利于其茎段增殖培养; IBA 0.2mg/L、6-BA 0.8mg/L、 KT 1mg/L组合下,叶绿素a、叶绿素b、叶绿素总含量及内源IAA、 GA、 ZT、 ABA含量、 IAA/ZR比值均达到最大; IBA对苗高生长、叶绿素总含量及内源ZR含量的影响较大,6-BA对茎段增殖培养及内源ABA含量的影响较大, KT对内源IAA、 GA含量影响较大。苗高与ABA/GA间呈极显著负相关,茎段增殖系数与IAA/ZR存在一定的负相关关系,但相关性均不显著;叶绿素总含量与ABA/ZR间呈极显著负相关。 ABA/GA比值的减少有利于苗高生长,而IAA/ZR比值的减少则有利于腋芽的诱导, ABA/ZR比值的减少能促进叶绿素的积累。%The effects of various plant growth regulators with different concentrations and ratios on seedling growth and endogenous hormones of Cunninghamia lanceolata tissue culture were studied based on L4 (23 ) orthogonal ex-perimental design. The results showed that different plant growth regulators could promote seedling height growth and stem growth of Cunninghamia lanceolata tissue culture seedling, the more suitable plant growth regulator con-centration for proliferation was IBA 0. 3 mg/L and 6-BA 0. 8 mg/L, the KT 1 mg/L was favorable for the seedlings growth, the KT 2 mg/L was favorable for the stem segment growth. The contents of chlorophyll a, chlorophyll b, total chlorophyll content, and the content of IAA, GA, ZT, ABA, and IAA/ZR ratio were all the maximum at the combination of IBA 0. 2 mg/L, 6-BA 0. 8 mg/L, KT 1 mg/L. The effect of IBA on the seedling height growth, chlorophyll content and the content of ZR was the highest, and the effect of 6-BA on the stem segment growth and the content of ABA was greater, and the effect of KT on the content of IAA and GA was higher. An ex-tremely significant negative correlation were found between seedling height and ABA/GA, the negative correlation between stem segment multiplication coefficient and IAA/ZR was not so significant, an extremely significant nega-tive correlation were found between total chlorophyll content and ABA/ZR. The decrease of the ratio of ABA/GA was favorable for the seedling height growth, and the decrease of the ratio of IAA/ZR was favorable for the induc-tion of axillary buds, and the decrease of the ratio of ABA/ZR was favorable for the accumulation of chlorophyll.
