热疗联合紫杉醇对动物接种肿瘤抑制作用的实验研究

         

摘要

目的:观察热疗联合紫杉醇对动物接种宫颈癌细胞系(U14)的抑制作用.方法:将U14瘤液接种在小鼠右后肢股外侧肌肉,待肿瘤平均瘤体积达150 mm3时,平均分组,根据实验要求,分别进行紫杉醇治疗、热疗、紫杉醇与热疗联合治疗和对照观察.当各组平均瘤体积≥2 000 mm3时,处死动物,取出肿瘤进行光、电镜检查.结果:当肿瘤平均体积为1 000 mm3和2 000 mm3时,联合治疗组的肿瘤生长延迟时间( TGD)为3d,q值分别为1.50和1.36.光镜观察:肿瘤细胞体积缩小,分布集中,肿瘤细胞变性坏死,血管壁增厚,数目较多,有肉芽形成,并有淋巴细胞浸润.电镜观察:大部分细胞膜系统有严重损伤,包膜模糊不清,包膜部分溶解,胞质外溢,线粒体内嵴断裂呈空泡状,溶酶体增多,粗面内质网(RER)有脱颗粒现象.结论:热疗与紫杉醇联合治疗对肿瘤组织显示协同杀伤作用,在临床治疗上具有应用价值.%Objective: To observe the effects of hyperthermia combined with paclitaxel in mice implanted cervical carcinoma cell line (U14). Methods: The suspension of tumor cell was inoculated subcutaneously in dorsum of the hind right foot of the experimental mice. When the average tumor volume rcached 150 mm3, they were divided into four groups: control group, paclitaxel administered group, hyperthermia group and the combined group (hyperthermia and paclitaxel). All of them were equally treated according to the experiment requirements. The tumor was observed by optical microscope. Results: In the combined group the tumor growth delay (TGD) was three days and the q value were 1.50 and 1.36 when the average tumor volume reached or exceeded 1000 mm3 and 2 000 mm3 respectively. Observations with optical microscope showed: the volume of tumor cell was reduced, distribution was concentrated, and tumor cell was necrosis in tissue; the thickness of vascular wall was increased, granulation tissue was formed; lymphacytes infiltration between skin and tumor tissue were found. Observations with electric microscope showed: the tumor cells membrane was destructed with partial dissolution; chondriosome was degenerated, lysoosome was increased; roughly-surfaced endoplasmic reticulum (RER) was degranulation. Conclusion: Combined therapy of hyperthermia and paclitaxel has a lethal synergic effect on tumor tissues provide a possibility for clinical practice.
