多效唑、矮壮素对彩色马蹄莲试管微型种球诱导的影响

         

摘要

[Objective]The present experiment was conducted to induce corm in test tube plantlet derived from Zantedeschia hybrida to enhance propagation efficiency and transplant survival rate, shorten seedling raising lime, reduce occurrence of serious disease and decrease high production cost. [ Method ]The test tube plantlets with 3-7 cm height and 0.3 cm diameter were used asexplants and inoculated to culture medium containing different sugar (2-8%), paelobutrazol (1 -8 mg/L) and chlorcholine chloride (2-16 mg/L), the formation rate of corm was calculated after culturing for 90 days. [ Result ]The formation rate of corm in Zantedeschia hybrida showed an increasing trend with increase in sugar in improved MS medium adding same paclobutrazol and chlorcholine chloride. With the increase of paclobutrazol and chlorcholine chloride in medium, the formation rate of corm increased initially, followed by a decrease. The induction effects of paclobutrazol (4 mg/L) and chlorcholine chloride (8 mg/L) was found the best. The highest corm formation rate was achieved in medium containing 4 mg/L paclobutrazol and 6% sugar. This combination also resulted in best average diameter and fresh weight of corm. The paclobutrazol and chlorcholine chloride had inhibitory effect on the growth of leaf, and differentiation and elongation of root [Conclusion]Adding suitable rate of sugar and paclobutrazol or chlorcholine chloride in improved MS+NAA (0.1 mg/L)+ agar (7 g/L) solid medium, can successfully induce the formation of corm in Zantedeschia hyhrida, and the induction effect of paclobutrazol was better than that of chlorcholine chloride.%[目的]利用植物生长调节物质对试管苗种球进行诱导,提高彩色马蹄莲种球繁育效率和组培苗移栽成活率,缩短育苗时间,减少病害发生,降低生产成本.[方法]挑选苗高3.0~7.0 cm、根茎直径0.3cm以上的试管苗,接人添加不同用量蔗糖(2%~8%)、多效唑(1~8 mg/L)、矮壮素(2~16 mg/L)的培养基培养90 d后统计成球率,筛选适合彩色马蹄莲试管微型种球诱导的培养基.[结果]在改良MS培养基中,在相同多效唑和矮壮素用量条件下,随着蔗糖用量的增加,彩色马蹄莲成球率呈上升趋势;随着多效唑、矮壮素浓度的升高,成球率均呈先上升后下降的趋势,分别以添加4 mg/L多效唑、8 mg/L矮壮素的诱导效果最好,其中以多效唑4 mg/L与6%蔗糖的处理组合成球率最高,达100%,且平均直径与鲜重最高,形成的种球最大(3.2 cm).多效唑与矮壮素对试管苗叶片、根的分化和伸长有明显的抑制作用.[结论]在改良MS+NAA 0.1 mg/L+琼脂7 g/L固体培养基中添加合适配比的蔗糖和PP333或多效唑,均可以成功诱导彩色马蹄莲组培苗在试管中结球,而多效唑对球茎的诱导形成作用明显优于矮壮素.
