不同穗型籼粳杂交稻F1代花药培养条件的优化

     

摘要

By using the F1 progenies of japonica and indica rice varieties SN265 / R99 with erect panicle and curved panicle as anther culture material, a study on different low -temperature pretrealment time, basic medium and concentrations and combinations of plant hormones was carried out. The result showed Low-temperature pretreatment at 10℃ for 7-11 days was to stimulate anther induction. Medium SK3 and M8 were better than the other media in callus induction. The 23.22 % of callus inducting rate in the SK3 medium added with 2 mg·L-1 2,4-D, 1 mg·L-1 NAA and 0.2 mg·L-1 6-BA was obtained and the 70.66 % of callus differentiation rate in the MS medium added with 4 mg·L-1 KT, 1 mg·L-1 NAA and 0.5 mg·L-1 6-BA were obtained.%以直立穗型沈农265粳稻与泸恢99籼稻杂交F1为材料,通过筛选与优化基本培养基种类、低温处理时间及激素成分等培养条件来建立高效花药培养体系.结果表明:经过7~11d的10℃低温处理的花药组织愈伤诱导显著高于未低温处理或其他处理时间的材料;以SK3或M8为基本培养基都可以获得较好的诱导率;以SK3为基本培养基,采用复合激素水平为2 mg·L-1 2,4-D +1 mg·L-1 NAA+ 0.2 mg·L-1 6-BA的愈伤组织诱导培养基获得最高23.22%的诱导率;以MS为基本培养基,复合激素水平为4mg·L-1 KT+1 mg·L-1 NAA+0.5mg·L-1 6-BA的分化培养基上获得最高70.66%的分化率.
