地塞米松对M-CSF体外诱导分化的骨髓巨噬细胞的影响

             

摘要

目的 研究地塞米松对单核细胞集落刺激因子(M-CSF)体外诱导分化的骨髓巨噬细胞的影响. 方法 M-CSF诱导分化的小鼠骨髓巨噬细胞,采用不同浓度地塞米松处理,进行形态学观察及增殖生长测定.免疫荧光法检测其吞噬功能,RT-PCR技术检测巨噬细胞各种细胞因子mRNA水平的表达. 结果 不同浓度地塞米松处理后,M-CSF体外诱导分化的骨髓巨噬细胞的增殖活性被明显抑制,且具有剂量依赖性;而且M-CSF体外诱导分化的骨髓巨噬细胞的吞噬活性被有效抑制.经过地塞米松处理后细胞趋于类圆形,伪足变宽、变短.地塞米松能抑制M-CSF体外诱导分化的骨髓巨噬细胞的促炎症因子mRNA水平的表达. 结论 地塞米松能有效抑制M-CSF体外诱导分化的骨髓巨噬细胞的活化及功能,为其作为免疫调节剂治疗噬血细胞综合征提供实验依据.%Objective To explore the regulatory effects of dexamethasone on bone marrow macrophage cultured by M-CSF in vitro. Methods M-CSF-induced bone marrow macrophage were cultured with the different concentrations of dexamethason.and the morphology and the cell proliferation were observed. Immunofluorescence method was performed to examine the phagocytic function,and RT-PCR technique was used to measure the mRNA levels of inflammatory factors. Results Proliferation of the cultured cells was inhibited by dexamethasone in a dose-dependent manner. The phagocytic functions of M-CSF-induced bone marrow macrophage were effectively inhibited by dexamethasone. The cultured cells were oval like,and their pseudopod became wide and short. Dexamethasone inhibited the related pro-inflammatory factors in the cultured cells. Conclusion The results suggest that dexamethasone could inhibit the activity and function of M-CSF-induced bone marrow macrophage in vitro,which provide an experimental basis in the treatment of hemophago-cytic lymphohistioeytosis.
