Caveolin-1和β-catenin在食管鳞癌中的表达及临床意义

         

摘要

Objective To explore the expression of Caveolin-1 and β-catenin in esophageal squamous cell carcinoma tissues (ESCC) and its relationships with invasion and metastasis.Methods The expression of Caveolin-1 and β-catenin in 110 specimens of esophageal squamous cell carcinoma tissues and 15 specimens of normal esophageal tissues was detected by immunohistochemistry technique.The relationship between their expression and clinicopathological features of ESCC were analyzed.Results The positive rate of Caveolin-1 in ESCC tissues was 73.64% (81/110),which was significantly higher than that in NEM tissues [33.33% (5/15),P <0.001].The positive rate of β-catenin in ESCC tissues was 82.73% (91/110),which was significantly higher than that in NEM tissues [20.00% (3/15),P < 0.001].The positive expression rates of Caveolin-1 and β-catenin were significantly higher in middle-low differentiation group than those in high differentiation group(P < 0.05).The positive expression rate of Caveolin-1 was higher in stage Ⅲ + Ⅳ group than those in stage Ⅰ + Ⅱ group(P < 0.05).The positive expression rates of Caveolin-1 were siguificantly higher in patients with lymph node metastasis or liver metastasis than those without metastasis (P < 0.05).There was no significant difference in positive expression of Caveolin-1 and β-catenin among different gender,age and tumor size.There was a positive con() between the positive expression of Caveolin-1 and β-catenin in MEC tissues (r =0.545,P =0.000).Conclusion The expression levels of Caveolin-1 and β-catenin are high in esophageal squamous cell carcinoma tissues,which are correlated with difrerentiation degree,TNM stage,lymph node metastasis,and liver metastasis.%目的 观察食管鳞癌(esophageal squamous cell carcinoma,ESCC)组织中癌基因Caveolin-1、β-catenin的表达变化,及其与肿瘤侵袭、转移的相关性. 方法 选取我院食管鳞癌组织110例,正常食管组织(normal esophageal mucosa,NEM)15例,分别采用免疫组化法检测两组食管组织的Caveolin-1、β-catenin的表达,分析Caveolin-1、β-catenin阳性表达与ESCC临床病理参数的关系及其在ESCC组织中的相关性. 结果 ESCC组Caveolin-1蛋白阳性表达率为73.64% (81/110),明显高于NEM组[33.33% (5/15)],差异有统计学意义(P<0.001);ESCC组β-catenin蛋白阳性表达率为82.73% (91/110),明显高于NEM组[20.00% (3/15)],差异有统计学意义(P<0.001);Caveolin-1和β-catenin在食管鳞癌中的表达呈正相关(r=0.545,P=0.000);ESCC组中低、中分化者Caveolin-1和β-catenin蛋白表达率明显高于高分化者,Ⅲ+Ⅳ期的阳性表达率明显高于Ⅰ+Ⅱ期,伴淋巴结转移、肝转移组的阳性表达率明显高于未转移者(P<0.05);而不同性别、年龄及肿瘤大小的Caveolin-1和β-catenin阳性表达率则无统计学差异(P>0.05);结论 食管鳞癌组织中Caveolin-1、β-catenin表达上调,其高表达与食管鳞癌分化程度、TNM分期、淋巴结转移及肝转移密切相关,二者具有协同作用.
