骨髓基质干细胞接种于珊瑚修复兔颅骨缺损的实验研究

         

摘要

Objective:To study the feasibility of natural coral (NC) granules loading osteogenesis in the repair of bone defects. Methods : Rabbit marrow stem cells(MSCs) were cultured and expanded in vitro . MSCs were seeded into NC granules at the deensity of 2×108/ml ,and cell-N C c omplexes were implanted into rabbit skull defects in the diameter of 15 mm.Cells and NC granules were implanted into the defects respectively as contro ls.Two months after operation, the animals were sacrificed and new bone regen e ration in the defects was investigated by gross specimen inspection, X-ray exam i nation and histological observation. Results:Two months after operation,most area of the NC-MSCs transplanted defects was filled with newly formed bone,and n ew bone integrated with NC granules.In N C granule implantated defects,bone regeneration was only noticed at the edge of h ost bone.In cell implantated defects,new bone could be observed in some central area of the defects. Conclusions:NC-MSCs may be feasible in the repair of bone defects.%目的:在骨缺损中植入骨形成细胞和细胞附着的支架材料,观察缺损的修复情况。方法:体外扩增兔骨髓基质干细胞(MSCs),将MSCs按2×108/ml的浓度与珊瑚(NC)颗粒复合后植入细胞供体兔颅骨直径15 mm缺损内,以单纯植入颗粒状珊瑚和MSCs作为对照。术后2月取材,通过大体标本观察、X线片检查、组织学检查观察骨缺损的修复情况。结果:MSCs-颗粒状NC 植入后2月,缺损大部分获得修复,新骨与NC颗粒相互融合,组织学观察可见缺损中有大量新骨形成;单纯MSCs植入后仅在缺损中央有新骨形成;单纯NC颗粒植入仅在缺损边缘有新骨形成。结论:在骨缺损中植入骨形成细胞和细胞附着的支架材料,可以明显提高骨缺损的修复效果。
