AGR2在卵巢上皮性癌患者血清及癌组织中的表达及意义

摘要

目的:探讨卵巢上皮性癌血清和癌组织中人前梯度蛋白2(AGR2)的表达水平与卵巢上皮性癌发生、发展的关系及用于其诊断的意义.方法:采用ELISA法和电化学发光法检测40例卵巢上皮性癌、10例卵巢交界性肿瘤、20例卵巢良性肿瘤及17例健康体检者血清AGR2、CA125水平；采用免疫组化法检测各种卵巢肿瘤组织及因宫颈癌切除卵巢的15例正常卵巢组织中AGR2的表达情况,并结合临床病理参数进行分析.结果:①卵巢上皮性癌及卵巢交界性肿瘤组织AGR2的阳性表达率明显高于卵巢良性肿瘤及正常卵巢组织(P ＜0.05,P＜0.01).②卵巢上皮性癌及卵巢交界性肿瘤血清中AGR2水平明显高于卵巢良性肿瘤及健康体检者(P＜0.05).③卵巢上皮性癌血清AGR2水平及癌组织中AGR2的阳性表达率均与临床分期及淋巴结转移相关.④联合检测血清AGR2和CA 125曲线下面积与单独检测CA125、AGR2比较,差异均无统计学意义(P均＜0.05).结论:AGR2与卵巢上皮性癌的发生、发展有关.联合检测血清AGR2和CA 125对卵巢上皮性癌的早期诊断有参考价值.%Objective:To detect the expression of Anterior gradient 2 (AGR2) in serum and tumor tissue of epithelial ovarian cancer patients, andinvestigate the potential correlation betweenthe occurrence and development of epithelial ovarian cancer and AGR2 expression. Methods;The serum levels of AGR2 and CA126 were determined by ELISA and chemiluminescent assay in 40 cases of ovarian epithelial cancer, 10 cases of borderline ovarian cancer,20 cases of benign ovarian cystadenoma and 17 cases of healthy women. Expressions of AGR2 protein in tissues were detected by immunohistochemisty. Results;The serum level of AGR2 in ovarian epithelial cancer was significantly higher than that in benign tumor and healthy women, ( P < 0. 05). The AGR2 protein expressions in ovarian epithelial cancer and bordline cancer were both significantly higher than that in benign tumor and normal ovarian. ( P<0. 05,and P<0. 01). The serum level of AGR2 and AGR2 protein expression in tissue are both correlated with the clinic stage and lymph node metastasis of ovarian epithelial cancer. The Area Under Curve (AUC)of combined detection of serum AGR2 and CA125 are significantly higher than detection of CA125 lonely. Conclusions :AGR2 may participate in the occurrence and development of epithelial ovarian cancer. The diagnostic sensitivity and specificity were both improved by combined detection of serum AGR2 and CA125.
