IFN-γ在高效抗转录病毒治疗AIDS患者中的免疫调节作用

         

摘要

目的:探讨辅助性 T 细胞1(Th1)分泌的γ-干扰素(IFN-γ)对获得性免疫缺陷综合症(AIDS)患者的免疫功能调节作用。方法:选取2011年1月至2014年1月收集的20例经实验室确诊的AIDS患者进行研究,AIDS接受了高效抗转录病毒治疗(HAART组)、选取20例健康自愿者作为健康组,分别于AIDS接受高效抗转录病毒治疗前、治疗6、12个月时分别采集研究对象的静脉血进行外周血单个核细胞(PBMC)分离,分为单独培养组和IFN-γ共培养组,检测培养液中细胞因子及免疫细胞的表达差异。结果:治疗前、治疗6、12个月,单独培养组、IFN-γ共培养组上清液中IL-12、IL-21的水平均显著地低于健康组(P <0.05);治疗6、12个月,IFN-γ共培养组上清液中IL-12、IL-21的水平均显著地高于单独培养组(P <0.05)。治疗前、治疗6个月、治疗12个月,单独培养组、IFN-γ共培养组IP-10的水平、CD4+CD25+Foxp3Treg 细胞表达水平均显著地高于健康组(P <0.05);治疗6、12个月,IFN-γ共培养组上清液中IP-10的水平均显著地高于单独培养组(P <0.05),IFN-γ共培养组上清液中CD4+CD25+Foxp3Treg细胞表达水平均显著地低于单独培养组(P <0.05)。结论:AIDS接受了高效抗转录病毒治疗能够改善患者的免疫水平,IFN-γ可以进一步刺激AIDS患者免疫细胞因子的分泌。%Objective To investigate the regulation of immune function in patients with acquired immune deficiency syndrome (AIDS) by the secretion of T cell 1 (Th1) gamma interferon gamma (IFN-γ) in patients with acquired immune deficiency syndrome. Methods From January 2011 to January 2014, 20 cases of laboratory-confirmed AIDS patients were collected for research. Patients with AIDS received highly active anti-retroviral therapy (HAART group), while 20 healthy volunteers were selected as control group. Before receiving highly active anti-retroviral treatment in six months and 12 months , the patients with AIDS were collected in blood study of peripheral blood mononuclear cells (PBMC) in separated culture group and IFN-γ co-culture group for testing culture differential expression of cytokines and immune cells in solution. Results Before treatment in 6 months and 12 months , the supernatants in the cultured alone group and IFN-γ co-culture group by IL-12, IL-21 levels were significantly lower than that in the healthy group (P < 0.05); After treatment in 6 months and 12 months , the supernatants in the IFN-γ co-culture group by IL-12 and IL-21 levels were significantly higher than that in the cultured alone group (PP < 0.05). Before treatment in 6 months and 12 months,the level of IP-10 and CD4 + CD25 + Foxp3Treg cells in the cultured alone group and IFN-γ co-culture group, were significantly higher than the healthy group (P < 0.05); After treatment in 6 months and 12 months, the supernatant in the IFN-γ co-culture group was significantly higher than the average in the cultured alone group in IP-10; CD4 + CD25 + Foxp3Treg cells in the IFN-γ co-culture group were significantly lower than that in the cultured alone group (P < 0.05). Conclusion Patients with AIDS received highly active anti-retroviral therapy can improve the immunity. IFN-γ may further stimulate the secretion of immune cytokine in patients with AIDS.
