电针预处理对老年大鼠术后认知功能的影响及机制研究

         

摘要

目的 研究电针预处理对老年大鼠术后认知功能的影响及其机制.方法 健康清洁的老年雄性SD大鼠60只,将大鼠随机分为3组(n=20):对照组(C组)、单纯手术组(S组)、手术+电针刺激组(E组).E组于术前电针刺激百会、内关、曲池及足三里,每天1次,连续刺激7d;S组和E组行肝脏缺血再灌注手术.分别于术前1d和术后1、3、5、7d进行水迷宫实验,记录逃避潜伏期时间和游泳距离.检测脑脊液中Aβ-42和磷酸化tau-181蛋白及海马组织中Wnt、β-连环素(β-catenin)及磷酸化糖原合成激酶-3β(p-GSK-3β)蛋白的表达.结果 与C组比较,S、E组术后第1~7天逃避潜伏期及游泳距离明显延长(P<0.05),S、E组Aβ及p-tau-181蛋白含量增加(P<0.05),S组Wnt蛋白、β-catenin蛋白及p-GSK-3β蛋白含量减少(P<0.05),E组Wnt蛋白、β-catenin蛋白及p-GSK-3β蛋白含量明显增加(P<0.05);与S组比较,E组术后第1~7天逃避潜伏期及游泳距离明显缩短(P<0.05),E组Aβ及p-tau-181蛋白含量减少(P<0.05),E组Wnt蛋白、β-catenin蛋白及p-GSK-3β蛋白含量明显增加(P<0.05).结论 预先给予电针刺激可以明显改善老年大鼠术后认知功能,其机制可能与激活Wnt/β-catenin信号通路有关.%Objective To study the effect of electroacupuncture pretreatment on cognitive function in aged rats and its possible mechanism. Methods 60 healthy male SD rats were randomly divided into 3 groups(n =20):control group (group C),simple operation group (S group),operation + electroacupuncture stimulation group (E Group),E group in the preoperative electroacupuncture stimulation Baihui、Neiguan、Qu pool and Zusanli,1 d,continuous stimulation 7 d. Group S and E were treated with hepatic ischemia-reperfusion. The water maze test (MWN)was performed at 1d before operation and 1,3,5 d and 7 d after operation,and the escape latency and swimming distance were recorded. The expression of Wnt protein ,β-catenin and phosphorylated glycogen synthase kinase-3β(p-GSK-3β) in cerebrospinal fluid were measured. Results Compared with group C ,the escape latency and swimming distance were significantly prolonged in group S and E at 1-7d(P<0.05),and the contents of Aβand p-tau-181 in S and E group were increased(P<0.05),the contents of Wnt protein,β-catenin protein and p-GSK-3β protein in group E were significantly increased (P < 0.05),and the contents of Wnt protein ,β-catenin and p-GSK-3βprotein in group S were significantly decreased(P<0.05);Compared with group S,the escape latency and swimming distance were significantly shortened in group E (P < 0.05),the levels of Aβ and p-tau-181 protein in group E were decreased(P<0.05),and Wnt protein andβ-catenin protein in group E were significantly lower than those in group E (P < 0.05),and p-GSK-3β protein were significantly increased (P <0.05). Conclusion Pretreatment with electroacupuncture can significantly improve the cognitive function of aged rats,the mechanism may be related to the activation of Wnt/β-catenin signaling pathway.
