Objective To analysis the genotype of varicella-zoster virus(VZV) which caused the varicella epidemic situation in Yingze, Taiyuan in 2013 for developing strategies of varicella prevention and control. Methods Specimens were collected from 23 children clinically diagnosed with varicella . Polymerase chain reaction (PCR) was performed to identify the VZV genome in the specimens. PCR was performed to amplify 447 bp fragment in the open reading frame (ORF) 22 followed by sequence analysis. Finally, the sequence analysis and cluster analysis were performed with Vector NTI Suite 6 and Clustalx programmes. Results There were 19 specimens which were positive for the wild VZV genome. The 447 bp fragments in the ORF 22 were amplified in 12 specimens. The comparison with reference strains sequence revealed that all the 12 positive specimens carried 6 key SNPs in the 447 bp fragment , which were consistent with J genotype. Conclusion The major genotype of VZV which caused the varicella epidemic situation is J genotype in Yingze , Taiyuan in 2013.%目的:分析太原市迎泽区2013年引发水痘疫情的水痘-带状疱疹病毒(VZV)流行株的基因型别,为预防控制水痘疫情提供依据。方法收集23份水痘患儿的临床标本。实时荧光定量聚合酶链反应(PCR)鉴定是否为VZV核酸阳性。PCR扩增447 bp大小的部分开放阅读框(ORF)22片段,并测序。用Vector NTI Suite 6和Clustalx软件分析核酸序列并与参考菌株进行聚类分析。结果19份临床标本鉴定为VZV野毒株核酸阳性,其中有12份标本的PCR产物在约447 bp处有明显的特异性条带。12条VZV的447 bp片段核酸序列的6个单核苷酸多态性关键位点与J基因型参考株完全一致。结论太原市迎泽区2013年引发水痘疫情的VZV主要流行株为J基因型野毒株。
