An enzyme activity assay in solid phase was developed to investigate the enzmye dynamics of CMCase and 0-glucosidase during the thermophilic phase of cattle manure-rice straw composting. The enzyme activities monitored in this study were able to reveal the dynamics of the activity of both cellulase and β-glucosidase could be attributed to the abiotic parameters of composting process. Furthermore β-glucosidase activity was found positively correlated with CMCase. The changes that occurred in a bacterial community by PCR-denaturing gradient gel electropho- reses(DGGEs)was analyzed, targeting the family 1 /3-glucosidase genes. The results showed that the structure of microbial community changed with composting time. The major bands were sequenced and compared with sequences obtained from ONA database by phylogenetic analysis.%以固相酶活试验方法检测牛粪-稻草堆肥高温期的CMC酶活性和β-葡萄糖苷水解酶活性,分析结果表明,两种酶活变化存在相关性,并受堆肥过程中的理化因素影响.设计简并引物,应用PCR-DGGE技术对β-葡萄糖苷水解醇GH1家族阳性的微生物群落的组成,及其时空分布进行研究,试验结果显示β-葡萄糖苷水解酶相关的功能性微生物群落在堆肥的进程中存在演替现象.
