猪PILRB基因的克隆及多态性分析

         

摘要

Ⅱ型配对免疫球蛋白样受体(Paired immunoglobulin-like type 2 receptors,PILRs)是近年发现的免疫球蛋白超家族成员之一,含有两个亚型,PILRα和PILRβ。在机体天然免疫反应中发挥重要作用,目前猪PILRB基因基因组序列尚未得到克隆、测序。研究利用PCR方法克隆猪PILRB基因部分基因组序列并分析该区域多态性。结果表明,获得猪PILRB基因序列长1127 bp,包含完整外显子1、内含子1、外显子2、内含子2、外显子3和部分内含子3。猪PILRB基因内含子1和内含子2为研究首次克隆,全长分别为9和314 bp。在该区域共检测到15个SNPs,其中6个为中度多态位点,共形成5种单倍体型,ACACCG为优势单倍体型。研究结果为进一步揭示这些SNPs对PILRβ功能的影响及其在猪抗病育种中的作用提供基础。%Paired immunoglobulin-like type 2 receptors (PILRs) composed of two subtypes, PILRαand PILRβ, are members of immunoglobulin superfamily found recently. They play important roles in host innate immune responses. However, genomic sequence of porcine PILRB gene has not been cloned and sequenced as yet. This study was designed to clone and analyze polymorphisms of partial genomic sequences of porcine PILRB gene by using PCR method. The sequence obtained was 1 127 bp in length and contained the complete sequence of exon 1, intron 1, exon 2, intron 2, exon 3 and partial of intron 3. Intron 1 and intron 2 of porcine PILRB gene, 9 and 314 bp in length, respectively, were first identified experimently. Total y, 15 SNPs were identified in the region. Six SNPs were medium polymorphic loci and group into five haplotypes of which ACACCG was dominant haplotype. The results provide a basis for further analyzing the effect of these SNPs on protein function, and the role in disease-resistant breeding as wel .
