Wistar 大鼠福氏佐剂性类风湿关节炎动物模型研究

     

摘要

①目的探讨 Wistar 大鼠佐剂性类风湿关节炎造模效果,为类风湿关节炎发生机制及治疗效果等相关研究中动物模型的应用提供实验依据。②方法将大鼠随机分为两组,分别为模型组和正常对照组,每组各20只。模型组于大鼠右足趾皮下注射福氏完全佐剂(CFA)0.15mL,造模后第7、14、21、28天对两组大鼠进行关节炎评分。测量每组大鼠右足趾关节肿胀度,分离脾脏并制备脾细胞悬液,计算脾细胞增殖率,ELISA 法检测血清中 IL-10与 IL-17的含量。③结果模型组致炎后7天足趾肿胀达峰值,14天后再度肿胀,21天出现皮肤溃疡,28天后溃疡面进一步扩大。大鼠模型组第14天脾细胞增殖率高于第7 天和第21天,差异有统计学意义(P <0.05),模型组不同时期脾细胞增殖率均高于对照组,差异有统计学意义(P <0.05);模型组第7、28天关节炎评分高于对照组,差异有统计学意义(P <0.05);第7、14、28天血清中 IL-10与 IL-17的含量高于对照组,差异有统计学意义(P <0.05)。④结论模型组关节肿胀明显,大鼠表现接近于人体的类风湿关节炎(RA),且造模过程简单、稳定,是研究 RA 的较理想模型。%Objective To explore the effect of adjuvant rheumatoid arthritis model in Wistar rats,and provide experimental evidence for the application of animal models in the study of the mech-anism of rheumatoid arthritis and the evaluation of therapeutic effect.Methods The rats were ran-domly divided into two groups,model group and normal control group,20 in each group.Rats in mod-el group were injected with CFA 0.15mL.Then respectively to arthritis score of each group rats, measure the joint swelling of rats in each group,and calculate the spleen cell proliferation rate at the 7,14,21 and 28 days.Meanwhile heart blood and serum was separated to detect the content of serum IL-10 and IL-17 with ELISA method.Results Rats in model group induced inflammation after about 7 days the right toe swelling reached the peak.Then the right toe swelling agained about 14 days.The skin ulcer in rats occurred around 21 days.At 28 days,further pathological changed and ulcer surface to further expanded.In model group,The 14 days rats’spleen cell proliferation rate was higher than 7 days and 21 days,and the difference was significant (P <0.05).The rats’spleen cell proliferation rate in model group was higher than that in control group at different times,and the difference was significant (P <0.05).The rats’arthritis score in model group at 7 days and 28 days was higher than the control group,and the difference was significant (P <0.05).The content of serum IL-10 and IL-17 at 7,14 and 28 days were higher than the control group,the difference was significant (P <0.05).Conclusion The rats’joint swelling of model group is obvious,and the performance of rats is closer to human RA.Then the modeling process is simple and stable,which is an ideal animal model of rheumatoid arthritis.
