Objective To investigate the expression of miR-21,miR-203a,miR-146a,miR-125b in different development stages of epidermal keratinocytes of psoriasis.Methods Clinical psoriasis lesion specimens in different periods were collected.Stem-loop was used to reverse transcription primers into cDNA.The expression of miR-203a,miR-21,miR-146a and miR-125b was detected by Real-time PCR of absolute quantification.Results Compared with normal skin tissues,miR-21,miR-203a,miR-146a was significantly up-regulated at progressive,silent and stationaryphase in psoriasis(P<0.05) while miR-125b was significantly down-regulated at the same phase in psoriasis (P<0.05).Conclusion MiR-203a,miR-21,miR-146a and miR-125b are closely related to the progress of psoriasis,but its mechanism of action in psoriasis need further verification by the target gene.%目的 探讨miR-21、miR-203a、miR-146a、miR-125b在银屑病不同发展时期表皮角质细胞的表达情况.方法 收集临床银屑病不同时期皮损标本,采用Stem-loop技术反转录miRNA,Real time PCR检测miRNA的表达,绝对定量方法计算miR-21、miR-203a、miR-146a、miR-125b的表达水平.结果 相比于正常皮肤组织,miR-21、miR-203a、miR-146a在银屑病的进行期、静止期、稳定期均呈现高水平表达(P<0.05),miR-125b则均呈现低水平表达(P<0.05).结论 miR-203a、miR-21、MiR-146a、miR-125b与银屑病进展密切相关,但是其作用机制还需要通过靶基因验证等进一步研究.
