TLR-2、TLR-4在强直性脊柱炎患者外周血单个核细胞中的表达及其临床意义

摘要

目的:探讨TLR-2和TLR-4在AS患者PBMC中的表达及其临床意义.方法:选择AS患者及健康志愿者各30例,分为AS组和健康对照组.采用RT-PCR及蛋白免疫印迹技术检测两组受检者PBMC中TLR-2和TLR-4的表达水平.结果:AS组TLR-2 mRNA、TLR-4 mRNA表达水平分别为1.59±1.43、0.60±0.73,均高于健康对照组的0.06±0.04、0.06±0.39(P均＜0.05).AS组TLR-4 蛋白表达水平为12.0±8.6,高于健康对照组的4.5±2.1(P＜0.05).AS患者的BASDAI、BASFI与TLR-2 mRNA表达水平呈正相关(r分别为 0.759、0.664,P＜0.01);AS患者的BASDAI与TLR-4 mRNA表达水平呈正相关(r=0.347,P＜0.01).结论:AS患者TLR-2、和TLR-4的表达水平高于健康志愿者,并与其自身疾病活动性存在一定程度的相关性,TLR-4或许可以作为评估AS病情活动的有价值的参考指标.%Objective: To explore the expression and significance of toll like receptor 2 (TLR-2) and toll like receptor 4 (TLR-4) in peripheral blood mononuclear cells (PBMC) in patients with ankylosing spondylitis (AS).Methods: Thirty patients with AS and 30 healthy volunteers were enrolled and assigned into AS group and control group, respectively.The expression of TLR-2 and TLR-4 in PBMC were determined by RT-PCR and western blot.Results: Significantly higher expression of TLR-2 mRNA, TLR-4 mRNA and TLR-4 protein was revealed in AS group than that in control group (1.59 ± 1.43 vs 0.06 ±0.039, 0.60 ±0.73 vs 0.06 ±0.39 and 12.0±8.6 vs 4.5 ± 2.1, respectively, P ＜ 0.05).BASDAI and BASFI were positively correlated with expression level of TLR-2 mRNA in AS group ( r1 = 0.759, r2 = 0.664, P ＜ 0.01).Meanwhile, BASDAI was significantly positively correlated with TLR-4 mRNA expression level in AS patients (r = 0.347, P ＜0.01).Conclusion: The expressions of TLR-2 and TLR-4 in AS patients are higher than those in controls, while the expression of TLR-2 and TLR-4 in AS patients is correlated with BASDAI and BASFI, suggesting TLR-4 a valuable assessment of disease activity in AS.
