养肺活血方对肺纤维化大鼠肺组织血管新生的影响

         

摘要

OBJECTIVE To investigate the influence of Yangfei Huoxue Fang on angiogenesis of PYM-induced pulmonary fibrosis in rats.METHODS Rats were punctured to the heart-side from tracheal cartilage ring gap after anesthesia and injected 5 mg/kg pingyangmycin(PYM)into trachea to build pulmonary fibrosis rats model and 5 mg/kg normal saline to build sham-operation group. The rats were gavage administration on the second day after operation. Sham-operation group and model group was given 5‰ sodium carboxymethyl cellulose sodium(CMC Na)solution,dexamethasone group was given dexamethasone distilled water with the same volume,and Yangfei Huoxue group was given Yangfei Huoxue Fang with the same volume. The rats were killed after anesthesia for gavage administration for 28 days. After Fixing left lung,embedding section and conventional dewaxing,we used immunohistochemistry SP to detect vascular endothelial growth factor(VEGF)of lung tissue and marked capillaries by CD34. Integral optical density(IOD)and microvascular density(MVD)were measured randomly and calculated by optical microscope and automatic image acquisition system. RESULTS VEGF of lung tissue in model group was significantly higher than that of sham-operation group(P<0.01),and that of Yangfei Huoxue group was significantly lower than that of model group(P<0.01). Microvessel density of lung tissue in model group was significantly lower than sham-operation group while that of Yangfei Huoxue group was significantly higher than model group. CONCLUSION Reducing VEGF level of lung tissue and increasing microvessel density may be one of the mechanisms of treating pulmonary fibrosis by Yangfei Huoxue Fang.%目的 探讨养肺活血方对平阳霉素诱导肺纤维化大鼠肺组织血管新生的影响.方法 麻醉大鼠后,经气管软骨环间隙向心端穿刺,气管内注入平阳霉素5 mg/kg建立大鼠肺纤维化模型,假手术组大鼠在同样麻醉条件下气管内注入等量生理盐水.术后第2天开始每天灌胃给药,假手术组和模型组予5‰羧甲基纤维素钠(CMC钠)溶液,地塞米松组予等容积地塞米松蒸馏水溶液,养肺活血组予等容积养肺活血方合剂,灌胃28 d,麻醉处死大鼠,将左肺固定,包埋切片,常规脱蜡,用免疫组化SP法检测肺组织血管内皮细胞生长因子(Vascular endothelial growth factor,VEGF)和CD34标记微血管.采用光学显微镜和全自动图像获取系统随机测量并计算平均光密度(IOD)值及微血管密度(Microvascular density,MVD).结果 模型组大鼠肺组织VEGF的水平明显高于假手术组(P<0.01),养肺活血组肺组织VEGF的水平明显低于模型组(P<0.01).模型组大鼠肺组织微血管密度明显低于假手术组,养肺活血组肺组织微血管密度明显高于模型组.结论 降低肺组织VEGF的水平和增加微血管密度可能是中药养肺活血方治疗肺纤维化的机制之一.
