斑蝥素、斑蝥素酸钾及斑蝥素酸钠作用胃癌细胞的研究

     

摘要

To investigate the inhibitory effect of cantharadin, potassium cantharidate and sodium cantharidate on human gastric carcinoma cell, human gastric carcinoma SG C7901 cell lines was cultured by cell culture technique, and MTT colorimetry was used to evaluate the inhibitory effect of cantharadin, potassium cantharidate and sodium cantharidate on cell growth in the present work. The tested concentrations of cantharadin were 10.20 μmol/L, 20.41 μmol/L, 30.61 μmol/L, 40.82 μmol/L, 51.02 μ mol/L, and that of potassium eantharidate or sodium cantharidate were 775 μmol/L, 15.50 μ mol/L, 23.26 μmol/L, 31.01 μuol/L and 38.76 μmol/L, or 8.85 μmol/L, 17.70 μ mol/L, 26.55 μmol/L, 35.40 μmol/L and 44.25 μmol/L, respectively. Cantharidin ( 10.20 μmol/L) displayed the inhibitory effect on the growth of SGC7901 cells, and was highly dosage - dependent with the IC50 value of 16.17 μmol/L. Also, 15.50 μmol/L potassium cantharidate and 17.70 μmol/L sodium cantharidate obviously demonstrated the inhibitory effect on the growth of SGC7901 cells, whose IC5o value were 20.41 μmol/L and 27.83 μmol/L, respectively. The results of assay suggested that cantharidin were more effective on gastric cancer inhibition compared with other two chemicals.%采用MTT法,研究芜菁体内斑蝥素、斑蝥素酸钾及斑蝥素酸钠对胃癌SGC7901细胞的抑制作用.抑制试验斑蝥素的浓度分别为10.20、20.41、30.61、40.82、51.02 μmol/L,斑螫素酸钾浓度分别为7.75、15.50、23.26、31.01、38.76 μmol/L,斑蝥素酸钠浓度分别为8.85、17.70、26.55、35.40、44.25 μmol/L.结果表明:斑蝥素浓度为10.20μmol/L时,对SGC7901细胞有显著抑制作用,且随药物浓度升高其抑制作用增强,呈剂量效应关系,抑制中浓度为16.17μmol/L;斑蝥素酸钾浓度为15.50 μmol/L时,对SGC7901细胞有显著抑制作用,且随药物浓度升高其抑制作用增强,呈剂量效应关系,抑制中浓度为20.41 μmol/L;斑蝥素酸钠浓度为17.70μmol/L时,对SGC7901细胞有显著抑制作用,且随药物浓度升高其抑制作用增强,呈剂量效应关系,抑制中浓度为27.83 μmol/L.斑蝥素酸钾和斑蝥素酸钠对胃癌SGC7901细胞的作用比斑蝥素差,提示它们对胃癌的治疗效果可能不如斑蝥素.
