诃子水提取物对肺癌A549细胞抑制作用的实验研究

摘要

目的:探讨诃子水提物对人肺癌细胞A549体外增值的影响和作用机制.方法:利用MTT法检测不同剂量组的诃子水提物对肺癌A549细胞增殖的影响,通过倒置显微镜观察肺癌A549细胞的形态学变化,流式细胞仪测定各浓度组诃子水提物对肺癌细胞A549作用后的周期变化.结果:诃子水提物对人肺癌细胞A549的增殖具有抑制作用,且有浓度和时间依赖性;在显微镜下观察到典型的凋亡细胞形态特征;流式方法测定的结果显示,诃子水提物阻滞肺癌A549细胞于G0/G1期.结论:诃子水提物对人肺癌细胞A549增殖具有抑制作用,其机制是诃子水提物使细胞周期阻滞于G0/G1期并诱导细胞凋亡.%Objective:To study the effect of chebula aqueous extract on A549 human lung cancer cells in vitro. Methods: MTT assay was used to test the effect of different doses of aqueous extract of Terminalia to lung cancer A549 cell proliferation, inverted microscope was used to observe the morphological changes of lung cancer A549 cells, flow cytometry was used to detect the effect of aqueous extract of Terminalia on A549 lung cancer cells cycle. Results; Chebula aqueous extraat can inhibit human lung cancer cell A549 proliferation and with concentration and time de-pendent. Microscopically, typical apoptotic morphology was observed. Water extract of Terminalia could block lung cancer A549 cells in G0/G1 phase. Conclusion; Chebula aqueous extract can inhibit human lung cancer cells A549 arrest cell cycle at G0/G1 phase and induce apoptosis.
