Toll样受体3与人微小病毒B19在桥本氏甲状腺炎发病机理中的作用

         

摘要

Objective:To explore the expression of TLR3 and B19 in Hashimoto's thyroiditis ( HT), in order to further clearly define the pathogenesis of HT. Methods: The immunohistochemical staining with streptavidin-perox-idase (SP) and immunoenzymatic double staining procedure were conducted to detect the expression and location of TLR3 and B19 in 33 tissue specimens from patients with HT. Results: Both TLR3 and B19 were distributed in thyroid follicular epithelial cells which were close to lymphoid follicles from HT patients and majority of their expression can be detected in the same location. We found that 18 samples of TLR3 positive IHC signals were from cytoplasm and 15 most positive IHC signals of TLR3 were from nuclei in HT specimens, meanwhile, 18 cases of B19 positive IHC signals were from cytoplasm, 11 B19 positive IHC signals were from nuclei and 4 cases were negative. Statistical analysis showed that the expression of TLR3 and B19 was positively correlated to each other ( r =0. 565,P =0.001). Conclusion; The co - expression of TLR3 and B19 can be detected in thyroid follicular cells from HT specimens and indicated that TLR3 can recognize B19dsRNA and at last lead to autoimmune diseases.%目的:探讨Toll样受体3(TLP3)和人微小病毒B19(B19)在桥本氏甲状腺炎(HT)发病机理中的作用,以进一步明确其发病机制.方法:选取33例桥本氏甲状腺炎患者的石蜡标本,分别采用免疫组化SP法及免疫组化双染法检测TLR3和B19在HT中的表达与定位,分析两者与HT发病的关系.结果:TLR3和B19主要表达在HT的甲状腺滤泡上皮细胞内,且大多数(62%)位于同一细胞内.在33例HT病例中,TLR3染色18例以胞浆表达为主,15例以胞核表达为主;B19染色18例主要表达于胞浆,11例主要表达于胞核,4例为阴性表达.统计学分析表明,TLR3和B19在HT中的表达具有相关性(r=0.565,P=0.001).结论:TLR3和B19在HT甲状腺滤泡上皮细胞呈共表达,表明TLR3可能通过特异性识别人微小病毒B19复制过程中产生的dsRNA,使促炎症因子及辅助刺激分子释放,引起自身免疫性疾病.
