Embelin对HL-60细胞MMP-9 mRNA、NF-κB及VEGF表达的影响

Objective: To investigate the effect of Embelin on matrix metalloproteinase - 9 ( MMP - 9) mRNA, vascular endothelial growth faclor( VEGF) and NF - KB expression in HL -60 cells and explore the relation between them. Methods: The expression of MMP -9 mRNA was detected by Real time -PCR, and VEGF and NF - KB by Western blot. Results: Embelin could inhibit the NF - KB.MMP -9 mRNA and VEGF expression in HL -60 cells in a dose -dependent manner. Conclusion; Embelin could downregulate MMP -9 mRNA,NF -KB and VEGF expression in HL - 60 cells and may reduce the invasiveness of leukemia cells.%目的:研究Embelin对人类髓系白血病HL-60细胞株基质金属蛋白酶-9(MMP-9)mRNA、血管内皮生长因子(VEGF)及NF-κB表达的影响,并探讨相互之间的关系.方法:应用RT-PCR检测MMP-9mRNA的表达,Western blot检测VEGF及NF-κB的表达.结果:Embelin可以抑制NF-κB、MMP-9 mRNA及VEGF的表达,且具有浓度依赖性.结论:Embelin可通过抑制NF-κB降低MMP-9 mRNA及VEGF表达,从而可能降低白血病细胞的侵袭性.
