miR197－p53途径在三阴性和激素受体依赖型乳腺癌中的研究

摘要

目的：探究miR197在三阴性和激素受体依赖型乳腺癌中的表达、作用机制以及预后。方法：收集健康女性（乳腺癌疑似患者活检后正常）乳腺样本（n＝10），三阴性乳腺癌患者（n＝40），激素受体依赖型[雌激素和（或）孕激受体阳性、人表皮生长因子受体－2（Her－2）阴性]乳腺癌患者（n＝40）的肿瘤样本。通过荧光实时定量PCR检测三组样本的miR197表达。通过蛋白免疫印迹法（Western Blotting），检测正常女性和不同类型乳腺癌患者p53以及凋亡通路蛋白。结果：低表达miR197占三阴性乳腺癌患者的77．5％（31／40）、占激素受体依赖型乳腺癌的42．5％（17／40）的和正常女性的20．0％（2／10），分别P＜0．05、P＜0．005。miR197表达高的细胞 p53的表达较低，低表达 miR197细胞 Caspase －3、Caspase －9表达增高，凋亡显著。结论：miR197－p53途径抑制细胞凋亡，与乳腺癌不良预后相关。%Objective:To investigate the expression and prognosis mechanism of miR197 in invasive breast canc-er.Methods:Collecting serum of healthy women(n=10),patients with hormone receptor dependent(ER or/and PR positive,Her-2 negative)breast cancer(n=40),triple patients with negative breast cancer(n=40)tumor samples. Detecting the miR197 in serum samples of three groups through the Real-time PCR.Detecting the expression of ap-optosis protein in two groups of patients with breast cancer by Western Blotting.Results:The low expression of miR197 in patients with 42.5% patients with triple negative breast cancer(31/40).Hormone receptor dependent breast cancer(17/40)and 20.0%normal women(2/10),P<0.05.miR197 expression in breast cancer patients with low expression of p53 was higher,and it was more likely to occur apoptosis.Conclusion:The expression of miR197 in breast cancer cells is low,and it is associated with poor prognosis.
