SPE-HPLC法测定杞菊地黄丸含莫诺苷和马钱苷的量

     

摘要

Objective To establish the SPE-HPLC method for detecting morroniside and loganin contents in Qijudihuang Pills. Methods The Eclipse XDB-C18 chromatographic column(250.0 mm×4.6 mm,5μm) was adopted,acetonitrile-0.100%phosphoric acid solution used as the mobile phase for conducting the gradient elution,the flow rate was 1.0 mL/min,the detection wavelength was 240 nm,the column temperature was 40℃,the sample size was 10μL. Results Morroniside and loganin showed good linear relation in the range of 0.050,2.000μg,the morroniside regression equation was Y=9.28×105X+14.4(r=0.999 2);the loganin regression equation was Y=1.02×105X+5.55(r=0.999 2),the average recovery rate was 102.835%and 99.080%respec-tively (RSD=1.400%,1.200%). Conclusion This method is simple,accurate and rapid,and can be used for the simultaneous determination of morroniside and loganin contents in Qijudihuang Pills to control the quality of preparation.%目的建立SPE-HPLC 法测定杞菊地黄丸含莫诺苷和马钱苷量的方法。方法采用 Eclipse XDB-C18(250.0 mm×4.6 mm,5μm)色谱柱,以乙腈-0.100%磷酸溶液为流动相进行梯度洗脱,流速为1.0 mL/min,检测波长为240 nm,柱温为40℃,进样量为10μL。结果莫诺苷和马钱苷分别在0.050、2.000μg范围内线性关系良好,莫诺苷的回归方程为 Y=9.28×105X+14.4(r=0.9992);马钱苷的回归方程为 Y=1.02×105X+5.55(r=0.9992),平均回收率分别为102.835%、99.080%[相对标准偏差(RSD)=1.400%、1.200%]。结论该法简便、准确、快速,可用于同时测定杞菊地黄丸中莫诺苷和马钱苷的含量,控制药品质量。
