1株三乙胺高效降解菌的筛选鉴定及其降解特性研究

             

摘要

A bacteria strain named as T-5 that could grow utilizing triethylamine as sole carbon source,nitrogen source,and energy was tamed and isolated from activated sludge in wastewater treatment pool in a pharmaceutical plant Shandong Province. It was an aerobic,Gram-positive,oxidase reaction positive,indole test reaction positive, lactose indecomposable,similarity of its 16S rDNA with Bacillus cereus was 99%,therefore,it was initially identified as Bacillus cereus based on 16S rRNA gene sequence analysis,homology comparison and analysis,morphological,and physiological properties. The pH,temperature and inoculation for the growth of T-5 were observed in shaker flash for the effect of its triethylamine degradable performance,the results showed that the optimal conditions were pH 7. 0,30℃,and 5%,respectively. T-5 was able to completely degrade 500 mg/L triethylamine within 48 hours under the op-timal degradable conditions. This research achievement has very important application value for efficient treatment of triethylamine pollution in the environment.%从山东某制药厂污水处理系统的活性污泥中驯化、筛选分离得到1株能够以三乙胺作为唯一碳源、氮源和能源进行生长的细菌,命名为T-5。根据菌落形态观察、生理生化特征和16S rDNA基因序列同源性对比分析对其进行鉴定,发现该菌为需氧、革兰阳性菌,氧化酶呈阳性反应,吲哚试验呈阴性反应,不分解乳糖,且该菌的16S rDNA序列与蜡状芽胞杆菌( Bacillus cereus)的相似度达99%,因此,初步鉴定该菌为蜡状芽胞杆菌。通过摇瓶实验考察了温度、pH、接种量对菌株T-5降解三乙胺性能的影响,实验结果表明该菌生长和降解三乙胺的最适培养条件为温度30℃,pH 7.0。在最适培养条件下,T-5能够在48 h内将500 mg/L三乙胺完全降解。研究成果对高效处理环境中的三乙胺污染具有重要的应用价值。
