银杏叶提取物对肝纤维化大鼠肝脏Smad2/3表达的影响

     

摘要

Objective To evaluate the effects of Ginkgo biloba extrat( GbE) on expression of Smad2/3 ,thus to investigate the possible mechanism of GbE in rats with hepatic fibrosis. Methods The liver fibrosis model was induced by CCl4. The rats in administration groups were given with 200mg/kg,100mg/kg,50mg/kg of GbE by daily gavage. At the end of the sixth week, all the rats were sacrificed and livers were obtained. The expression of Smad2/3 was detected by immunohistochemistry. The expression of Smad2/3mRNA in liver was measured hy hybridization in situ. Results The expreasion of Smad2/3mRNA and Smad2/3 protein were significantly reduced after the administration of GbE and Fufang Biejia Ruangan Pian in model group. Conclusion GbE could reduce the transcription and expression of Smad2/3 , and inhibit the occurrence and development of CCl4 - induced liver fibrosis.%目的 观察银杏叶提取物(GbE)对肝组织Smad2/3表达的影响,探讨GbE抗肝纤维化的作用机制.方法 采用CCl4诱导大鼠肝纤维化,同时分别以200、100、50mg/(kg·d)的GbE进行干预,并设复方鳖甲软肝片为对照组,6周后取肝组织,原位杂交及免疫组化技术分别检测肝组织Smad2/3mRNA及蛋白的表达.结果 模型组大鼠肝组织Smad2/3mRNA及蛋白的表达正常组明显增高,应用不同剂量的GbE及复方鳖甲软肝片干预后肝组织Smad2/3mRNA及蛋白较模型组显著下降.结论 GbE降低Smad2/3mRNA及蛋白表达可能是其抗肝纤维化作用的重要机制之一.
