胰岛素信号通路相关基因变异与2型糖尿病关系

     

摘要

胰岛素分泌缺陷和胰岛素抵抗是2型糖尿病(type 2 diabetes mellitus,T2DM)发病的2个重要环节,胰岛素的敏感性和胰岛素分泌缺陷都具有家族聚集现象,提示可以通过研究T2DM的中间环节,阐述T2DM的发病机制.文中就最新发现的全基因组扫描获得的与T2DM相关的胰岛素信号通路系统中重要的基因如胰岛素受体底物1(insulin receptor substrate,IRS-1)、PTPRD、SRR的变异与2型糖尿病的关系作一综述,探讨其作为药物治疗的新靶点及对患者个体化防治的意义.%Insulin secretion deficiency and insulin resistance are two important factors in the pathogenesis of type-2 diabetes mellitus ( T2DM ). Both insulin sensitivity and insulin secretion deficiency have a family aggregation phenomenon, which suggests that the pathogenesis of T2DM could be expounded by studying the intermediate links in T2DM. This article summarizes the relationship of T2DM with the variation of such important insulin receptor substrates as IRS-1, PTPRD and SRR in the insulin signaling pathway, and evaluates this new target of drug therapy in individualized prevention and treatment of T2DM.
