3T动态磁共振磷谱对正常人骨骼肌线粒体功能的在体研究

     

摘要

目的:运用3T高场强磁共振动态磷谱技术(<'31>P-MRS)在体评价骨骼肌线粒体能量代谢情况.方法:对20名正常人受试者骨骼肌进行动态<'31>P-MRS采集,后期利用Matlab软件对无机磷(Pi)、磷酸肌酸(PCr)、三磷酸腺苷(ATP)等化合物的峰下面积进行定量分析,分别计算在静止期,运动末期及数个恢复期骨骼肌内高能磷酸化合物的含量,同时计算二磷酸垛苷(ADP)和细胞内PH值,评价磁共振动态磷谱技术对研究骨骼肌线粒体功能的价值.结果:肌肉运动时PCr含量明显降低,Pi、Pi/PCr和ADP升高,恢复期各含磷化合物含量逐渐恢复至静息水平.结论:3T高场强动态磷谱技术可以无创性定量评价骨骼肌线粒体功能,为肌肉的功能影像学提供客观证据,为以后客观研究肌肉相关疾病提供了理想的方法.%Objective: To evaluate the skeletal muscle mitochondrial function using in vivo dynamic phosphorus magnetic resonance spectrascopy (31P-MRS) on clinical 3T MR system. Methods : 31 P-MRS was acquired from skeletal muscles in 20 healthy volunteers. spectra were analyzed using madab software. Concentration of inorganic phosphate (Pi) , phosphocreatine (PCr) and adenosine triphosphate ( ATP) of every state were measured from area under the peak separately. Concentration of adenosine diphosphate (ADP) and intracellular pH value were calculated from the biochemistry reaction equilibrium. Results :During exercise, PCr concentration decreased, Pi, Pi/PCr and ADP values increased, in recovery state. Concentration of every phosphate compound come back to the resting state gradually. Concluslon: 3 T high field intensity dynamic phosphorus magnetic resonance spectroscopy technique can rellect skeletal muscle mitochondrial function in vivo and provide valuable information to evaluate muscle functional imaging, thereby representing an ideal solution to research muscles related diseases.
