乌桕优选单株的离体快速繁殖技术研究

             

摘要

选择8年生乌桕优树母株,取其枝条的腋芽茎段为外植体,进行组织培养快速繁殖技术研究。结果表明,接种时,先用70%的酒精对带芽茎段消毒30s,再用0.1%升汞灭菌11min,污染率可降到18.5%。在初代培养中,芽诱导最佳培养基为WPM+6-BA1.5mg/L+NAA0.2mg/L,30d后的诱导率可达85.4%。在4/5MS+6-BA1.5mg/L+KT0.5mg/L+NAA0.05mg/L培养基中继代增殖培养,增殖效果最好,增殖倍数为3.8,生长高度达4.8cm。将健壮的试管芽苗接种于1/2MS+IBA0.5mg/L+NAA0.3mg/L+Citricacid100mg/L培养基上进行生根诱导,生根率可达95.8%。移栽35d后,成活率达87.8%。%The stem sections with axillary bud of Sapium sebiferum ( L. ) Roxb. ( 8 years old ) were used as explants for the research of tissue culture and rapid propagation. The results showed that the contamination rate was 18.5% when we steri- lized the explants with ethanol(70% ) 30 seconds then used HgC1 z (0. 1% ) to sterilize the stem sections 11 minutes. The medium suitable for induction in primary culture was WPM + 6 - BA 1.5 mg/L + NAA 0.2 rag/L, and the inductivity reached to 85.4% after 30 days. The medium suitable for secondary proliferation was 4/5 MS + 6 - BA 1.5 mg/L + KT 0. 5 mg/L + NAA 0. 05 mg/L. The proliferation coefficient was 3.8 and the bud length could reach to 4. 8 cm. When the sturdy test-tube plantlets were inoculated into the medium of 1/2 MS + IBA 0.5 mg/L + NAA 0. 3 mg/L + Citric acid 100 mg/L for rooting induction, the rooting rate became over 95.8%. The transolantin~ survival rate was 87.8% after 35 days.
