阿司匹林对心肌肥厚大鼠左心室重构的影响及其与炎症因子的关系

         

摘要

Objective: Study the aspirin influence on pressure overload myocardial hypertrophy structure of rats, and the relationship of inflammatory markers TNF-αand IL-10.Methods: 54 rats were randomly grouping, Groups of rats were measured at 4 wk and 8 wk after surgery, calculation of arterial blood pressure (LVWI). Using HE staining and Masson staining to observe the changes of myocardial collagen fibers.Using ELISA method to detect myocardial and inflammatory cytokines in plasma TNF alpha, IL-10 levels. Results: Postoperative abdominal aorta 4 wk and 8 wk, Blood pressure in the model group and LVWI increased significantly in the control group (P < 0.05). The ventricular structure's changes.During each group rat in two time points 4 wk and 8 wk, plasma's TNF alpha, the content of IL-10 no significant difference (P> 0.05); In the Model 4 wk and 8 wk group, the TNF alpha content in myocardial tissue significantly increased (P<0.05).With the development of the process of myocardial hypertrophy, the content of anti-inflammatory factor IL-10 in myocardial tissue significantly increased (P<0.05), While application of aspirin can reduce pressure overload rats myocardial tissue TNF alpha and IL-10 levels (P<0.05); Conclusion: The experimental results show that the reticular fiber reduce.Inflammatory factor to promote the development of myocardial hypertrophy.Aspirin have anti-inflammatory effect, which can effectively reduce the expression of inflammatory cytokines, has certain protective effect.%目的:研究阿司匹林对压力超负荷性心肌肥厚大鼠左心室结构的影响,及其与炎症因子TNF-α、IL-10的关系.方法:将54只Wistar大鼠随机分为假手术4wk组、假手术8wk组,模型4wk组、模型8wk组、阿司匹林+模型4wk组和阿司匹林+模型8wk组.各组分别测动脉血压、计算左心室质量指数(LVWI)、HE染色观察心肌组织结构、Masson染色观察心肌胶原纤维的改变.用ELISA法检测心肌和血浆中炎症因子TNF-α、IL-10含量.结果:腹主动脉缩窄术后4wk和8wk,模型组动脉血压及LVWI较假手术组明显升高(P<0.05);模型4wk和8wk组心肌细胞肥大明显,胶原纤维增生;模型4wk和8wk组心肌组织中TNF-α含量显著升高(P<0.05);模型8wk组心肌组织中抗炎因子IL-10含量也显著升高(P<0.05),而应用阿司匹林可以降低压力超负荷大鼠心肌组织中TNF-α和IL-10的含量显著降低(P<0.05).结论:在心肌肥厚发生早期,即出现心肌细胞肥大,胶原纤维增生的变化;炎症因子促进了心肌肥厚的发展;阿司匹林具有抗炎作用,可有效降低炎症因子的表达,对于预防心肌肥厚的发生有一定的保护作用.
