补肾化瘀方对PCOS大鼠卵巢颗粒细胞CYP450表达的影响

摘要

目的 现察补肾化疼方对多囊卵巢综合征(PCOS)大鼠卵巢颗粒细胞CYP450蛋白表达的影响.方法 运用补肾化瘀方对PCOS模型大鼠进行干预后,采用免疫组化法观察卵巢颗粒细胞CYP450的蛋白表达.结果 模型组与正常组比较,卵巢颗粒细胞CYP450的蛋白表达在表达面积及表达强度上均低于正常组,差异具有统计学意义(P＜0.01);补肾化瘀方中、高、低剂量组及妈富隆组用药后,卵巢颗粒细胞的GYP450的蛋白表达明显升高,差异具有统计学意义(P＜0.01,P＜0.05);其中补肾化瘀方中荆量对PCOS大鼠卵巢颗粒细胞CYP450的蛋白表达效果最佳.结论 补肾化瘀方能增强PCOS大鼠卵巢颗粒细胞CYP450的蛋白表达,调节卵巢内分泌失调,改善PCOS高雄血症,逆转卵泡的发育异常,使排卵成功.%Objective To explore the effects of Bushen Huayu decoction (BHD) on protein expression of CYP450 in ovarian granulosa cells in PCOS rats.Method BHD was used to interpose the rats and the protein expression of CYP450 was determined with immunohistochemical method.Results Compared with normal group, the protein expression of CYP450 in model group was lower (P＜0.01); and in high, middle and low dosages of BHD groups and western medicine group were obviously higher(P＜0.01 or P＜0.05), in which, in middle dosage of BHD group was best.Conclusion BHD can increase the protein expression of CYP450 in PCOS rats, regulate the maladjustment of incretion balance and improve hyperandrogenemia, restore the abnormal growth of folliole and ensure the success of ovulation.
