动物组织中苯并咪唑类药物检测的LC- MS/MS法研究

     

摘要

建立了可食性动物组织中阿苯达唑、阿苯达唑-2-氨基砜、阿苯达唑亚矾、阿苯达唑砜、噻苯咪唑、5-羟基噻苯咪唑、奥芬达唑砜等7种苯并咪唑类药物及其代谢物的多残留检测的高效液相色谱-串联质谱法.分析过程中以保留时问和2个MRM离子对(母离子和2个子离子)进行定性,以峰形好和响应值高的离子进行定量.结果表明,该方法检测苯并咪唑类药物残留的检测限为2 ng/mL,定量限为10 ng/mL.建立的方法灵敏、可靠,7种苯并咪唑类药物在2.0~500 ng/mL范围内具有良好的线性,准确度和精密度都能满足残留检测的要求,可作为动源性食品中苯并咪唑类药物残留的定量检测和确证方法.%A liquid chromatographic/tandem mass spectrometric (LC/MS/MS) multiresidue method for the simultaneous determination of 7 benzimidazoles and metabolites residues in muscle and liver was established in this paper. Benzimidazoles and metabolites were extracted from tissues by ethyl acetate,purified by hexane and waters MCX cartridge. The LC separation was performed on a reversed-phase C18 column using gradient elution. The benzimidazoles and metabolites analytes were detected with a quadrupole mass spectrometer using positive ion electrospray ionisation. MS data acquisition was generally performed in the multiple reaction monitoring (MRM) mode with the mobile phase of water-acetonitrile-formic acid. The limit of detection (LOD) of the method was 2 ng/mL and the limit of quantification (LOQ) was 10 ng/mL.
