不同类型分子标记在苜蓿基因组中的检测效率分析

         

摘要

In order to select the suitable molecular marker for detection of alfalfa genome,19 domestic and foreign alfalfa germplasm as materials,the detection efficiency in alfalfa genome was compared for several types of molecular markers including ISSR, SCoT and RAPD. Results showed that there was great difference among the different types of molecular markers. The higher screening ratio of primer was found for RAPD, and it reached 41. 18%. The higher number of total loci and polymorphic loci were also detected for RAPD. ISSR had the higher percentage of polymorphic loci,and its percentage of polymorphic loci reached 96. 25%. For the effective allele number,Nei's gene diversity index,Shannon's information index,SCoT performed better than other types of molecular markers. These results demonstrated that the different molecular markers had advantages and disadvantages in different detection index, and these molecular markers should be used comprehensively.%为选择适合苜蓿基因组检测的分子标记,以19份国内外苜蓿种质为材料,比较分析了ISSR、SCoT、RAPD 3种标记在苜蓿基因组中的检测效率.结果表明,不同分子标记检测效率差异较大,引物筛选率以RAPD标记最高,达到41.18%;扩增总条带数和多态性条带数均以RAPD标记最多,多态性条带百分比以ISSR最高,为96.25%;SCoT标记的有效等位基因数、Nei's基因多样性指数和Shannon's信息指数值均高于ISSR、RAPD标记.可见,不同分子标记在不同检测指标方面各有优劣,应综合应用.
