远志水解产物的分离及鉴定

             

摘要

Objective To isolate and identify the hydrolysis product of radix polygalae, to elucidate rule of hydrolization initially. Methods Polygalae was extracted by diluted alcohol, was enriched by n - butanol and macroporous resin. The total saponin was hydrolyzed by hydrochloric acid.The separation was performed on silica gel column by high performance liquid chromatography ( HPLC ) .The structure of compounds were identified through ultraviolet (UV) , nuclear magnetic resonance ( NMR) , mass spectrometry ( MS) . Results Four compounds from hydrolysis product of Radix polygalae were identified, polygalic acid ( 1 ) , senegenin (2) , polygalic acid - 3 - O - β - D - glucoside (3 ) and senegenin -3 - O - β - D - glucoside (4). Conclusion This method is efficient to obtain polygalacic acid and provide a basis to improve the quality standard of radix polygalae further.%目的 对远志药材水解产物进行分离,鉴定,初步探讨其水解规律.方法 远志药材用稀醇提取,正丁醇萃取,大孔吸附树脂富集得到总皂苷,总皂苷酸水解后,用硅胶色谱柱、制备液相方法 分离得单体化合物,利用紫外光谱、核磁共振、质谱法等鉴定技术,确定单体化合物结构.结果 从远志皂苷水解产物中分离得到了4个单体化合物,分别为,远志酸(1),远志皂苷元(2),远志酸-3-O-β-D-葡萄糖苷(3),远志皂苷元-3-O-β-D-葡萄糖苷(4),其中3、4为新化合物.结论 发现了一条成熟的高效制备远志酸对照品的路线,为进一步完善远志质量标准奠定了基础.
