CpG序列诱发抗dsDNA抗体的免疫学特性分析

         

摘要

Objective To investigate the immunochemical propertyies of anti - dsDNA antibodies induced in normal mice by immunization with a cytosine guanine dinucleotides ( CpG) motif as adjuvant in normal mice. Methods Using oligonucleotide containing CpG motif as an adjuvant, native calf thymus DNA was used as the mimic self antigen to immunize normal BALB/c mice. After immunization, the specificity and immunochemical properties of anti - DNA antibodies induced in normal mice were analyzed. Results The induced and spontaneous anti - DNA antibodies had a similar ranges in avidity and binding kinetics. The induced antibodies displayed a predominance of IgG2a which could bind to DNA from three different species. Conclusion The CpG motif can promote normal mice to produce anti - dsDNA antibodies that resemble those in spontaneous systemic lupus erythematosus.%目的 研究胞嘧啶鸟嘌呤二核苷酸(cytosine guanine dinucleotides,CpG)序列诱生抗dsDNA抗体的免疫特征.方法 以CpG序列为生物佐剂,用天然的小牛胸腺DNA模拟自身抗原,免疫正常的BALB/c小鼠.免疫结束后,分析小鼠血清中抗dsDNA抗体的特征.结果 CpG序列诱生的抗dsDNA抗体类似自发性狼疮小鼠体内的自身抗体,主要为IgG2a型,且能与多种DNA抗原反应.结论 CpG序列能促进正常小鼠产生类似自发性狼疮小鼠体内的抗dsDNA抗体.
