低分子肝素对人乳腺癌细胞MCF-7侵袭抑制实验研究

             

摘要

目的 探讨低分子肝素(low molecular weight heparin,LMWH)抑制人乳腺癌细胞MCF-7的侵袭作用及其机制.方法 采用四甲基偶氮唑蓝比色法检测LMWH素对人乳腺癌细胞MCF-7黏附的影响,采用Transwell小室法检测LMWH对该细胞的迁移、侵袭的抑制作用,采用逆转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)法半定量检测LMWH处理细胞24h后组织因子(tissue factor,TF)mRNA的表达水平.结果 MTT比色法分析检测显示,LMWH对MCF-7细胞与细胞外基质成分Matrigel黏附力的抑制作用呈剂量效应依赖关系.迁移和侵袭实验显示,LMWH对MCF-7细胞的迁移和侵袭抑制均呈剂量-效应依赖关系.半定量RT-PCR检测结果显示,LMWH处理后MCF-7细胞TFmRNA表达水平较对照组明显降低.结论 LMWH可通过下调TFmRNA表达水平抑制人乳腺癌细胞MCF-7黏附、迁移、侵袭.%Objective To study the effects of low molecular weight heparin( LMWH ) on invasiveness of human breast cancer cell line MCF-7 and its mechanisms. Methods Tetrzolium-based colorimetric assay ( MTT ) colorimetric method was used to detect the effect of LMWH on the adhesion of human breast cancer cellline MCF-7. The expression of tissue factor ( TF ) mRNA was semiquantitatively detected by reverse transcription polymerase chain reaction ( RT-PCR ) in MCF-7 cells after being treated with LMWH for 24h. Results LMWH inhibited the adhesion of MCF-7 cells significantly in a dose-dependent manner. LMWH inhibited the invasive and metastatic abilities of MCF-7 cells significantly in a dose-dependent manner. The results of RT-PCR showed that the expression of TFmRNA decreased after LMWH treatment for 24h compared to that of control group. Conclusion LMWH inhibited the adhesive, metastatic and invasive abilities of MCF-7 cells by down-regulating the expression of TFmRNA.
